NAC类转录因子在‘砀山酥梨’黄化叶复绿过程中的表达分析

摘  要:为探讨NAC转录因子在梨缺铁黄化叶复绿过程中的表达特性,筛选响应该过程的核心NAC因子,以清水处理正常梨树叶片(N)和黄化梨树叶片(C)为对照(C_(N)和C_(C)),探讨外源0.2%FeSO_(4)溶液对梨缺铁黄化叶Fe^(2+)积累的影响,研究其关键PbrNAC基因的生物学性质、组织表达特异性以及复绿过程中的表达模式,阐述其与叶内Fe^(2+)积累的关联性。结果表明,(1)于N和C内共鉴定到含NAM结构域的21个显著差异表达的PbrNAC基因,涉及8个亚族,其长度不均,motif数从4~9不等,内含子数较少;(2)该基因倾向于根中表达,且均可响应缺铁黄化叶的复绿,其中Pbr032231.1、Pbr021393.1、Pbr026635.1、Pbr038615.1、Pbr019210.3、Pbr019212.1、Pbr002372.1这7个基因在C_(C)内的表达量显著低于C_(N)内表达量,且FeSO_(4)处理后,各时期表达量均较C_(C)显著上调;与之相反,Pbr007284.1、Pbr016205.1、Pbr007673.1等14个PbrNAC基因均在C_(C)内显著高表达,而FeSO_(4)处理后,各时期表达量总体较C_(C)显著下降;(3)各叶样内Fe^(2+)含量与Pbr007284.1、Pbr016205.1、Pbr007673.1基因的表达量均呈显著负相关,且该3个基因表达与Pbr016932.1、Pbr027956.1、Pbr029956.1、Pbr026635.1等基因表达显著相关;(4)多数PbrNAC在正常梨树根系和黄化梨树根系中的表达趋势与其在C_(N)和C_(C)内的表达趋势基本一致,说明其在地下部根系与地上部叶内可能存在表达协同性。以上结果表明,梨PbrNAC基因可能与梨树缺铁胁迫响应和缺铁黄化叶的复绿相关,其中Pbr007284.1、Pbr016205.1、Pbr007673.1可能起重要调控作用。To explore the roles of NAC transcription factors(TFs)in the progress of the retrieved green of Fe-deficiency chlorotic leaves in‘Dangshansuli’(Pyrus bretschneideri Rehd.),PbrNAC TFs were screened in the pear leaves,and the expression patterns of key PbrNACs in response to the retrieved green progress were analyzed.In this study,normal leaves(N)and chlorotic leaves(C)of‘Dangshansuli’were taken as test material,and exogenous 0.2%FeSO_(4)(2 g/L FeSO_(4))solution was sprayed on C during the growing season to confirm its effect on Fe2+accumulation in C,with N and C treated with distilled water as the control(C_(N)and C_(C)).The authors studied the biological properties,tissue-specific expressions,and response patterns of the key PbrNACs in this progress.Moreover,the correlation analysis between the expressions of PbrNACs with Fe2+content was carried out.The results showed,(1)A total of 21 PbrNAC genes with typical NAM domain were significantly differently expressed between N and C,which could be clustered into 7 subclasses with fewer introns and with motifs ranging from 4 to 9.(2)Most of the PbrNAC genes preferred to express in the roots,and all the genes could respond to the retrieved green progress of C.Among them,the expression levels of the seven genes named Pbr032231.1,Pbr021393.1,Pbr026635.1,Pbr038615.1,Pbr019210.3,Pbr019212.1,Pbr002372.1 was significantly lower in C_(C)than that in C_(N),and after FeSO_(4)treatment,their expressions were all significantly increased compared to that of C_(C).In contrast,the expression levels of 14 PbrNAC genes,including Pbr007284.1,Pbr016205.1,Pbr007673.1,were notably higher in C_(C)than that in C_(N),and after FeSO_(4)treatment,they were more like-ly to increase with statistical differences.(3)The Fe2+content in leaves was significantly negatively asso-ciated with the expressions of Pbr007284.1,Pbr016205.1,and Pbr007673.1.Although other NAC TFs did not show a direct linkage with the Fe2+accumulation,there was a certain relationship between them and Pbr007284.1,Pbr01620
