摘 要:目的探讨卵巢子宫内膜异位症(EMs)是否发生了上皮-间质转化(EMT)现象,以及miR-200b在EMs中的表达情况及在EMs发生发展中的作用。方法采用实时荧光定量聚合酶链反应(RT-PCR)技术检测EMs患者异位子宫内膜(ECs)和在位子宫内膜(EUs)以及正常子宫内膜(ENs)中miR-200b的表达水平。采用Western blot法检测所有标本中Snail蛋白的表达水平。采用非参数Student t检验或方差分析(ANOVA)进行统计分析,然后进行两两多重比较。结果在不同内膜组织中,miR-200b有不同程度的表达。与ENs组织相比,ECs和EUs组织中的miR-200b表达水平明显降低。并且ECs中miR-200b表达水平较EUs组织下调。相反,与ENs组织相比,ECs和EUs组织中Snail表达量明显升高,且ECs组织中的Snail表达水平明显高于EUs组织。Pearson相关系数检验显示,miR-200b和Snail呈显著的负相关。结论在卵巢EMs中,miR-200b表达水平降低,下调的miR-200b可能通过降低对EMT转录因子Snail的抑制作用从而促进Snail的表达,从而促使EUs组织发生了EMT,增加了EMs的发病风险。Objective To explore whether epithelial-mesenchymal transition(EMT)involved in ovarian endometriosis(EMs),and to investigate the expression of miR-200b in EMs and its role in the occurrence and development of EMs.Methods The expression levels of miR-200b was quantified using real time fluorescent quantitative polymerase chain reaction(RT-PCR)in ectopic endometrium(ECs)and eutopic endometrium(EUs)in patients with EMs and normal endometrium(ENs)in EMs-free subjects.The expression levels of Snail protein in all samples were evaluated by Western blot analysis.Statistical analysis was performed by nonparametric Student t test or analysis of variance(ANOVA)followed by post hoc pairwise multiple comparisons.Results MiR-200b in each tissue has different degree expression.Compared with ENs tissue,the expression level of miR-200b in ECs and EUs were decreased obviously.Moreover,the expression of miR-200b in ECs was down-regulated compared with that in EUs.On the contrary,the expressions of Snail increased significantly in ECs and EUs tissues compared with ENs tissues,and the expressions of Snail were significantly higher in ECs than EUs.According to Pearson Correlation Coefficients test,miR-200b had a significantly negative correlation with Snail.Conclusion The expression of miR-200b is obviously decreased in ovarian EMs.Down-regulated miR-200b may promote Snail expression by reducing the inhibition of EMT transcription factor Snail,thus promoting the EMT in EUs,and increasing the risk of EMs.
