基于16SrDNA技术研究针刺对气郁质肠道菌群的调节机制

摘　　要：目的:基于16SrDNA技术研究针刺对气郁质肠道菌群的调节机制。方法:招募气郁质和平和质受试者各20例。对气郁质受试者进行针刺干预8周,每周3次。观察干预前后的气郁质评分、17项汉密尔顿抑郁量表评分(HAMD)、汉密尔顿焦虑量表(HAMA)评分变化,并采集受试者新鲜粪便进行16SrDNA测序分析。结果:针刺可显著降低气郁质、HAMD及HAMA评分(P<0.05),提升气郁质受试者菌群丰度并调节Beta多样性;气郁质评分与瘤球胃菌属及粪杆菌属呈正相关,与小杆菌属呈负相关;针刺干预后,气郁质受试者瘤球胃菌属减少,克雷伯氏菌属升高(P<0.05),且肠道菌群氧化应激耐受能力和需氧菌丰度显著提升。结论:针刺可改善气郁质受试者临床症状,其作用机制可能与调节肠道菌群结构及肠道内氧化环境有关。Objective:To study the regulation mechanism of acupuncture on the gut microbiota of qi stagnation constitution based on 16S rDNA technology.Methods:Twenty participants with qi-stagnation constitution received acupuncture intervention for 8 weeks,while 20 participants with a balanced constitution did not receive any intervention.The score of qi stagnation constitution,HAMD and HAMA were used to evaluate the change of clinical characteristics after acupuncture,and the fresh feces of the subjects were collected for 16S rDNA sequencing.Results:Acupuncture could significantly reduce qi stagnation constitution,HAMD and HAMA scores(P<0.05),improve gut microbiota abundance in participants with qistagnation constitution,and regulated their gut microbiota Beta diversity.Qi stagnation constitution score was positively correlated with ruminococcus and faecalibacterium,and was significantly negatively correlated with Dialister;acupuncture decreased the Ruminococcus and increased the klebsiella,in addition,acupuncture significantly improved the oxidative stress of the gut microbiota(P<0.05),and the abundance of aerobic bacteria in qi stagnation constitution.Conclusion:Acupuncture is beneficial to the clinical symptoms of subjects in qi stagnation constitution,and its mechanism may be related to the regulation of gut microbiota structure and intestinal oxidative environment.
