摘  要:为探索甜菜雄性不育系与保持系在形成过程中的差异基因,以甜菜不育系(MS137,MS301)和保持系(OT154,OT302)为材料,取二年生花蕾进行转录组测序,经分析共获得花蕾期差异基因257个,GO功能注释发现差异基因主要富集于催化活性、水解活性、生物过程中的碳水化合物代谢及转运。KEGG功能结果注释表明差异基因涉及新陈代谢、戊糖及葡萄糖醛酸转换及淀粉和蔗糖代谢。结果表明有8个显著差异表达基因可能参与甜菜育性调控,其中,GDSL酯酶基因、半乳糖醛酸酶基因、果胶裂解酶基因、果胶酯酶基因等在不育系中显著下调表达,相关基因表达量的降低,花粉成熟过程受阻,形成不育;UDP-葡糖基转移酶基因、细胞色素P450基因在不育系显著上调表达,植物生长素类分泌过剩,导致花粉败育,甜菜雄性不育。研究结果为进一步探索甜菜不育系育性调控机理提供理论依据。Transcriptome sequenc ing was performed on biennial flower buds from male sterile lines(MS137,MS301) and maintainer lines(OT154,OT302) of sugar beet.A total of 257 differentially expressed genes at flower bud stage were obtained by analysis.GO functional annotation showed that differentially expressed genes were mainly concentrated in catalytic activity,hydrolytic activity,carbohydrate metabolism and transport in biological processes.KEGG functional annotation indicated that the differentially expressed genes were involved in metabolism,pentose and glucuronic acid conversion,and starch and sucrose metabolism.Based on GO and KEGG analysis of differential expression,8 differentially expressed genes were selected,including SGNH hydrolase gene,GDSL esterase gene,galacturonase gene,pectin lyase gene,pectin esterase gene and glycoside hydrolase,which were all up-regulated in CMS lines.UDP-glucosyltransferase gene and cytochrome P450 gene were up-regulated in maintainer lines.The results provide a theoretical basis for further exploring the mechanism of fertility regulation of male sterile lines in sugar beet.
