摘  要:为探究人为噪音对瘤背石磺(Onchidium reevesii)氧化应激和免疫应答的影响,将瘤背石磺分别暴露在1000 Hz的人为噪音中1、3、6、12和24 h后,分别测定瘤背石磺血清中超氧化歧化酶(SOD)、过氧化氢酶(CAT)等氧化应激指标和谷草转氨酶(AST)、谷丙转氨酶(ALT)活性和碱性磷酸酶(AKP)等免疫指标,同时测定了肝胰腺中热休克蛋白27(Hsp27)、谷胱甘肽过氧化物酶(GSH-Px)、谷胱甘肽巯基转移酶(GST)、髓样分化因子(MyD88)和肿瘤坏死因子-α(TNF-α)等基因的mRNA表达量变化。结果显示,相对于未暴露组,SOD和CAT活性在噪音暴露1 h后显著升高(P<0.05),AST和ALT活性在噪音暴露3 h后显著升高(P<0.05),AKP活性在噪音暴露1 h后显著升高(P<0.05);Hsp27和GSH-Px基因的mRNA表达量在噪音暴露1 h后显著上调(P<0.05),GST基因的mRNA表达量在噪音暴露1 h后显著下调(P<0.05),MyD88和TNF-α基因的mRNA表达量在噪音暴露1 h后显著上调。研究表明,人为噪音作为一种胁迫因子,使瘤背石磺机体产生了氧化应激和免疫应答,研究结果可为表征潮间带动物应对人为噪声胁迫提供参考依据。In recent years,anthropogenic noise has emerged as a significant source of pollution,posing a serious threat to the health and survival of marine species.While previous studies have mainly focused on the effects of anthropogenic noise on the growth,behavior,and auditory system of marine organisms,limited research has been conducted on physiological changes,which are closely linked to the health,reproduction,and survival of marine animals.In this study,we investigated the effects of anthropogenic noise on oxidative stress and immune responses in Onchidium reevesii.O.reevesii was exposed to anthropogenic noise at 1000 Hz and 80-108 dB for varying durations(1,3,6,12,24 h).We assessed changes in serum oxidative stress indicators,including superoxide dismutase(SOD)using the WST-1 assay and catalase(CAT)using the ammonium molybdate spectrophotometric method.Furthermore,immune indicators,such as glutamate transaminase(AST)and alanine transaminase(ALT)were measured using the Reitman Frankel assay,and alkaline phosphatase(AKP)levels were determined using disodium phenyl phosphate.Quantitative real-time PCR was utilized to analyze the mRNA expression levels of oxidative stress-related genes,such as heat shock protein 27(Hsp27),glutathione peroxidase(GSH-Px),and glutathione mercaptotransferase(GST),as well as immune response-related genes like myeloid differentiation factor(MyD88)and tumor necrosis factor-α(TNF-α),in hepatopantras.Results showed that,compared with O.reevesii in the unexposed group,SOD and CAT activities significantly increased after 1 h of exposure(P<0.05),reaching their maximum at 12 h,while AST and ALT activities significantly increased after 3 h of exposure(P<0.05),reaching their maximum at 6 h.Additionally,AKP activity significantly increased after 1 h of exposure(P<0.05),reaching its maximum at 12 h.The mRNA expressions of Hsp27 and GSH-Px genes significantly increased after 1 h of exposure(P<0.05),reaching their maximum at 12 h.In contrast,the mRNA expression of GST gene significantly decreased afte
