夏桑菊浸膏对高渗诱导干眼模型人角膜上皮细胞保护机制研究

摘  要:目的研究夏桑菊浸膏对高渗诱导下人角膜上皮细胞的保护作用及机制。方法采用NaCl构建高渗模型,通过MTS法检测细胞活力;利用RT-qPCR及酶联免疫吸附测定法检测相关炎症因子IL^(-1)β、IL-6、IL^(-1)8的表达;2′,7′-二氯荧光黄双乙酸盐荧光探针法检测细胞内活性氧水平。结果对比对照组,模型组600 mOsm·L^(-1)中细胞活力明显下降,经夏桑菊浸膏处理后,细胞活力明显上升;在炎症指标中,与对照组相比,模型组450 mOsm·L^(-1)细胞中IL^(-1)β、IL-6及IL^(-1)8的分泌水平与IL^(-1)β表达水平明显升高。夏桑菊浸膏处理细胞后,以上指标较450 mOsm·L^(-1)模型组明显下降;抗氧化研究中,模型组450 mOsm·L^(-1)中的细胞活性氧水平明显上升,夏桑菊浸膏处理后,细胞活性氧水平明显降低。结论夏桑菊浸膏对高渗诱导下的人角膜上皮细胞活力下降起到抑制作用,在高渗环境下具有保护人角膜上皮细胞的能力,保护作用通过抑制炎症因子产生和活性氧水平升高实现。Objective To investigate the protective effect and mechanism of the extract of Xiasangju on human corneal epithelial cells induced by hypertonic.Methods Hypertonic model was constructed with NaCl,and cell viability was detected by MTS method.The expressions of IL^(-1)β,IL-6 and IL^(-1)8 were detected by RT-qPCR and enzyme-linked immunosorbent assay.Intracellular reactive oxygen species(ROS)were detected by 2′,7′-dichlorofluorescein yellow diacetate fluorescence probe.Results Compared with the control group,the cell vitality of the model group at 600 mOsm·L^(-1)was significantly decreased,and the cell vitality of the model group was significantly increased after treatment with the extract of Xiasangju.In terms of inflammatory indicators,the secretion levels of IL^(-1)β,IL-6 and IL^(-1)8 and the expression levels of IL^(-1)βin 450 mOsm·L^(-1)cells in the model group were significantly increased compared with the control group.Compared with 450 mOsm·L^(-1)model group,the above indexes were significantly decreased after treatment with the extract of Xiasangju.In the antioxidant study,the reactive oxygen species in 450 mOsm·L^(-1)cells in the model group was significantly increased,while the reactive oxygen species was significantly decreased after treatment with Xiasangju extract.Conclusion The extract of Xiasangju can inhibit the decrease of human corneal epithelial cell vitality induced by hypertonic,and can protect human corneal epithelial cells in hypertonic environment by inhibiting the production of inflammatory factors and inhibiting the increase of reactive oxygen species.
