摘  要:目的:对比乙腈和甲醇作为流动相对多西他赛浓度测定实验的影响。方法:建立多西他赛浓度测定的色谱方法,分为实验组和对照组,实验组的流动相为乙腈,对照组的流动相为甲醇,两组除流动相外,其余色谱条件均保持一致,分别从专属性、标准曲线、定量下限、精密度、回收率、稳定性和柱压方面对实验组和对照组进行全方位比较。结果:实验组多西他赛和内标紫杉醇的保留时间分别为6.5 min和4.2 min左右多西他赛和内标紫杉醇的保留时间分别为10.3 min和8.1 min左右,两组的专属性均满足要求;实验组和对照组中多西他赛在0.03~10μg·mL^(-1)线性关系明确,两组多西他赛的定量下限均为0.03μg·mL^(-1);实验组在不同浓度的日内精密度为4.50%、5.40%和6.10%,日间精密度为9.80%、10.30%和10.70%,对照组在不同浓度的日内精密度为5.10%、7.60%和6.80%,日间精密度为12.50%、11.60%和13.10%;实验组在不同浓度的提取回收率为93.11±6.02%、93.32±3.24%和92.97±3.88%,对照组在不同浓度的提取回收率为90.34±5.67%、92.89±3.66%和90.77±3.79%;实验组(RSD)在不同浓度的RSD为9.3%、7.1%和6.3%,对照组在不同浓度的RSD为9.5%、6.8%和6.1%;实验组的柱压在相同流速下低于对照组。结论:乙腈对比甲醇更适合作为多西他赛浓度测定的流动相。Objective:To compare the effects of acetonitrile and methanol as flow relative to docetaxel concentration determination experiments.Methods:The chromatographic method for the determination of docetaxel concentration was established and divided into experimental group and control group.The mobile phase of the experimental group was acetonitrile,and the mobile phase of the control group was methanol.The chromatographic conditions of the two groups were consistent except for the mobile phase.Results:The retention time of docetaxel and internal standard paclitaxel in the experimental group was about 6.5min and 4.2 min,respectively,and the retention time of docetaxel and internal standard paclitaxel was about 10.3 min and 8.1 min,respectively.The specificity of both groups met the requirements.The linear relationship of docetaxel in the experimental group and the control group was clear in the range of 0.03~10μg·mL^(-1),and the lower limit of quantitation of Docetaxel in both groups was 0.03μg·mL^(-1).The intra-day precision of the experimental group was 4.50%,5.40%and 6.10%at different concentrations,and the intra-day precision was 9.80%,10.30%and 10.70%;the control group was 5.10%,7.60%and 6.80%at different concentrations,and the intra-day precision was 12.50%,11.60%and 13.10%.The recovery rates of the experimental group were 93.11±6.2%,93.32±3.24%and 92.97±3.88%at different concentrations,while the recovery rates of the control group were 90.34±5.67%,92.89±3.66%and 90.77±3.79%at different concentrations.The RSDS of the experimental group were 9.3%,7.1%and 6.3%at different concentrations,and those of the control group were 9.5%,6.8%and 6.1%at different concentrations.The column pressure of the experimental group was lower than that of the control group at the same flow velocity.Conclusion:Acetonitrile is more suitable as mobile phase for determination of docetaxel concentration than methanol.
