摘 要:目的:皮层注射氯化亚铁溶液建立外伤性癫痫大鼠模型及高压氧治疗对其防治及作用机制。方法:采用大脑皮层注射氯化亚铁建立外伤性癫痫大鼠模型,分模型组、治疗组及正常组进行行为学观察及脑电图监测,取脑组织观察神经细胞形态改变,检测各组样本c-fos mRNA、c-jun mRNA、c-myc mRNA的表达水平。结果:皮层注射氯化亚铁建立外伤性癫痫大鼠模型,操作简单,成功率高,稳定可靠,是值得推广的全新造模方法。该模型中c-fos、c-jun、c-myc均呈现长时程表达。结论:皮层注射氯化亚铁可通过诱导神经细胞凋亡导致癫痫发作。高压氧治疗外伤性癫痫可通过阻断c-jun、c-myc基因介导的细胞凋亡通路而起到治疗效果。Objective:Methods of FeCl_(2)modeling in rats with posttraumatic epilepsy and the prevention and mechanism of action of hyperbaric oxygen therapy.Methods:A rat model of posttraumatic epilepsy was established by injecting FeCl_(2)into the cerebral cortex,and the model group,treatment group and normal group were divided for behavioral observation and electroencephalogram monitoring,and brain tissues were taken to observe the morphologic changes of neuronal cells,and to detect the expression levels of c-fos mRNA,c-jun mRNA,and c-myc mRNA in samples from each group.Results:Cortical injection of FeCl_(2)to establish a rat model of posttraumatic epilepsy is simple,successful,stable and reliable,and is a new modeling method worth promoting.In this model,c-fos,c-jun and c-myc showed long time-course expression.Conclusion:Cortical injections of FeCl_(2)may cause seizures by inducing apoptosis in neuronal cells.Hyperbaric oxygen therapy for traumatic epilepsy can be therapeutic by blocking the c-jun and c-myc gene-mediated apoptotic pathway.
