摘 要:目的:研究徐长卿洗剂对体外马拉色菌的抑制作用及抗炎作用,进一步探讨其对马拉色菌感染的脂溢性皮炎的治疗作用。方法:测定徐长卿洗剂的最低杀菌浓度(MFC)并通过扫描电镜观察药物处理后的马拉色菌形态;通过脂多糖(LPS)诱导RAW264.7细胞炎症因子上调,分别利用荧光定量PCR(qRT-PCR)及酶联免疫吸附剂(ELISA)在转录水平及翻译水平测定徐长卿洗剂对肿瘤坏死因子(TNF-α)及白细胞介素-6(IL-6)表达的影响;建立皮肤感染马拉色菌的豚鼠动物模型,检测徐长卿洗剂对马拉色菌感染、皮肤损伤及机体免疫的干预作用。结果:实验结果表明徐长卿洗剂对马拉色菌生长具有显著的抑制作用,其MFC为1.25 mg/mL,扫描电镜观察发现徐长卿洗剂具有破坏马拉色菌菌体的功能。体外培养的RAW264.7细胞抗炎实验发现,徐长卿洗剂处理可以明显抑制LPS诱导升高的TNF-α(P<0.01)、IL-6(P<0.01)水平。豚鼠皮肤感染马拉色菌的治疗结果发现,与模型组相比,徐长卿洗剂能够促进马拉色菌诱导的皮损组织修复,同时通过恢复Th1、Th2细胞中白细胞介素-2(IL-2)(P<0.05)、干扰素γ(INF-γ)(P<0.05)、白细胞介素-4(IL-4)(P<0.05)、白细胞介素-10(IL-10)(P<0.05)表达增强机体免疫。结论:徐长卿洗剂可以有效治疗马拉色菌感染的脂溢性皮炎,其机制可能与调节Th1/Th2细胞平衡有关。Objective:To investigate the inhibitory and antiinflammatory effects of Vincetoxicum pycnostelma Lotion on Malassezia,and further explore the therapeutic effect of V.pycnostelma Lotion on Malassezia infected seborrheic dermatitis model.Methods:The minimal fungicidal concentration(MFC)of V.pycnostelma Lotion was detected and the morphology of Malassezia was observed by scanning electron microscope after treatment.Inducing upregulation of inflammatory factors in RAW264.7 cells through lipopolysaccharide(LPS),and using qRT-PCR and enzyme linked immunosorbent assay(ELISA)experiments to determine the effects of V.pycnostelma Lotion on tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)at transcriptional and translational levels.The effects of anti-bacteria,skin wound repair and immunoregulation of V.pycnostelma Lotion on the model of guinea pig infected with Malassezia were evaluated.Results:V.pycnostelma Lotion showed inhibitory effect on the growth of Malassezia,and the MFC was 1.25 mg/mL.Scanning electron microscope result revealed V.pycnostelma Lotion had the ability to destroy the structure of Malassezia.Then,the levels of TNF-α(P<0.01)and IL-6(P<0.01)in LPS-induced RAW264.7 cells were down-regulated significantly by V.pycnostelma Lotion.Finally,in comparison with model group,V.pycnostelma Lotion could repair skin damage and enhance host immunity by restore the levels of interleukin-2(P<0.05),interferon-γ(P<0.05),interleukin-4(P<0.05)and interleukin-10(P<0.05)secreted by Th1 and Th2 cells on the model of Malassezia infected guinea pigs.Conclusion:V.pycnostelma Lotion displayed therapeutic effect on Malassezia infected seborrheic dermatitis,and the underlying mechanism may be related with regulating Th1/Th2 cell balance.
