摘  要:目的比较不同配比高良姜-干姜提取物对胃寒的干预效果,为探讨高良姜-干姜提取物调理胃肠功能的药效研究提供依据。方法采用冰水加NaOH灌胃法建造胃寒证模型小鼠,将小鼠随机分为9组(n=10):正常组、模型组、吗丁啉组、附子理中丸组、干姜组、高良姜组以及低、中、高剂量高良姜+干姜组(简称联合低、中、高剂量组),持续给药30 d,1次/d。观察并记录小鼠舌象、腹部温度和身体质量的变化;计算小鼠胃内残留率、小肠推进率和各脏器指数变化;ELISA法测定胃组织的胃泌素(GAS)、胃动素(MTL)、乙酰胆碱(ACh)、超氧化物歧化酶(SOD)、丙二醛(MDA)以及白介素6(IL-6)的水平;观察各组小鼠胃组织的病理形态。结果与正常组比较,模型组小鼠胃组织病理呈现胃炎症状,胃组织中GAS、MTL、ACh和SOD水平显著降低,MDA和IL-6水平显著升高(P<0.01);待给药后,与模型组比较,联合低、中、高剂量组小鼠胃组织病理呈现胃炎症状减轻,舌象呈现淡红色,有光泽;小鼠胃组织中GAS、MTL、ACh和SOD水平、小肠推进率和胃排空率、体温显著升高及身体质量增高,MDA和IL-6的水平则显著下降(P<0.01或0.05)。结论高良姜+干姜联合给药可显著改善胃寒模型小鼠的胃寒症状,提高小鼠体温,促进小鼠胃肠蠕动能力,可能与其上调GAS、MTL、ACh、SOD水平以及下调MDA和IL-6水平有关。Objective To compare therapeutic effects of galangal-dried ginger extract of different proportions on stomach cold and provide evidence for the pharmacodynamic study on galangal-dried ginger extract in regulating gastrointestinal function.Methods The gastric cold syndrome model mice were established by intragastric administration of ice water and NaOH.The mice were randomly divided into 9 groups(n=10):Normal Group,Model Group,Motilium Group,Fuzi Lizhong Pill Group,Dried Ginger Group,Galangal Group and High-,Medium-and Low-Dose Galangal+Dried Ginger Groups(hereinafter referred to as High-,Medium-and Low-Dose Combination Groups).All the mice received continuous administration for 30 d,once a day.The changes of tongue manifestation,abdominal temperature and body weight of mice were observed and recorded.The gastric residual rate,intestinal propulsion rate and organ indexes of mice were calculated.The levels of gastrin(GAS),motilin(MTL),acetylcholine(ACh),superoxide dismutase(SOD),malondialdehyde(MDA)and interleukin 6(IL-6)in gastric tissue were measured by ELISA.The pathological morphology of gastric tissues in each group was observed.Results Compared with the Normal Group,the gastric histology of the Model Group presented gastritis symptoms,and the levels of GAS,MTL,ACh and SOD in the gastric tissue of the Model Group were significantly decreased,the levels of MDA and IL-6 of the Model Group were significantly increased(P<0.01).After administration,compared with the Model Group,the gastric histopathology of High-,Medium-and Low-Dose Combination Groups showed alleviated gastritis symptoms,the tongue manifestation of High-,Medium-and Low-Dose Combination Groups showed faint red and glossy,the levels of GAS,MTL,ACh and SOD,the intestinal propulsion rate and gastric evacuation rate,body temperature and body mass level of High,Medium-and Low-Dose Combination Groups were increased,and IL-6 levels of High-,Medium-and Low-Dose Combination Groups decreased significantly(P<0.05 or 0.01).Conclusion Combined medication of gala
