摘 要:目的通过超声对川崎病(KD)小鼠模型冠状动脉和心脏功能的测量及冠状动脉不同阶段病理分析,为KD模型的制备提供方法学证据。方法制备干酪乳杆菌细胞壁成分(LCWE)终液,培养50只BALB/c小鼠,将其分为3组,分别为模型组、治疗组及对照组;模型组和治疗组小鼠腹膜内注射LCWE终液建模,治疗组于实验第5天腹腔注射丙种球蛋白(IVIG),对照组小鼠腹腔内只注射同剂量生理盐水。采用小鼠心脏超声心动图分别于造模第0、第7、第15、第21、第30天获取的冠状动脉内径,记录M型超声并计算心脏功能;每次超声检查后均处死1只模型组小鼠以获取其冠状动脉病理。结果模型组左右冠状动脉直径在第15、第21及第30天大于治疗组及对照组。M型超声检查结果显示,造模后第15、第21及第30天模型组小鼠心脏功能低于治疗组及对照组(P<0.05);病理检查结果显示,造模后可出现冠状动脉内膜纤维结缔组织弥漫增生、管壁增厚、冠状动脉弹力纤维不连续等病理结果。结论利用LCWE诱导KD小鼠模型,通过高分辨率小动物心脏超声动态观察小鼠冠状动脉内径与心脏功能变化,可以用来模拟KD冠状动脉损伤及心功能的变化过程。Objective To provide methodological evidence for the preparation of Kawasaki Disease(KD)model,the coronary artery and cardiac function at different stages of KD mouse model were measured by ultrasound,and the pathology of coronary artery was analyzed.Methods Lactobacillus casei cell wall extract(LCWE)was prepared,50 BALB/c mouse were cultured and divided into three groups,namely the model group,the treatment group and the control group,The model group and the treatment group mouse were injected with LCWE terminal solution intraperitoneally,then the intravenous immune globulin(IVIG)was injected 5 days after modeling in the treatment group,only the same dose of normal saline was injected intraperitoneally in the control group of mice.The inner diameter of coronary arteries were measured by murine echocardiography on 0,7,15,21 and 30 days of the modeling,and the M-mode ultrasound was recorded to cardiac function calculation.One of the model group was sacrificed after every echocardiography for coronary pathology.Results The left and right coronary artery diameters in the model group were larger than treatment group and control group on 15,21 and 30 days after modeling.M-mode ultrasound showed that the cardiac function of the model group was lower than treatment group and the control group on 15,21 and 30 days after modeling(P<0.05).The pathological results showed that dffuse hyperplasia in fibrous connective tissue of intima,wall thickening in coronary artery,discontinuity in coronary arterial elastic fiber and so on could occur after modeling.Conclusions The changes in inner diameter of coronary artery and cardiac function in LCWE-induced KD mouse model can be observed dynamically by high-resolution small animal heart ultrasound,this model could be used to simulate the change of coronary artery injury and cardiac function of KD.
