摘  要:本试验旨在探讨灌服构树叶多糖(BPP)是否可以缓解氧化鱼油饲粮对大鼠肝脏健康的负面影响,并初步探讨其可能机制。选取32只健康状况良好的21日龄雄性断奶Wistar大鼠(平均体重为58.05 g),采用2×2析因设计,试验因素为灌服BPP[灌服BPP或磷酸盐缓冲溶液(PBS)]和氧化鱼油攻毒(饲喂氧化鱼油饲粮或正常鱼油饲粮),每日BPP的灌服量为22.5 mg/kg BW,氧化鱼油攻毒于试验第9天开始进行,试验期为26 d。结果显示:氧化鱼油攻毒提高了大鼠的腹泻率,显著提高了大鼠肝脏和血清中谷草转氨酶与谷丙转氨酶活性及血清中尿素氮含量(P<0.05),显著上调了大鼠肝脏中半胱氨酸天冬氨酸蛋白酶3和Kelch样环氧氯丙烷相关蛋白-1的mRNA相对表达量及B淋巴细胞瘤-2相关X蛋白与B淋巴细胞瘤-2的mRNA相对表达量的比值(P<0.05),显著下调了大鼠肝脏中B淋巴细胞瘤-2的mRNA相对表达量(P<0.05),显著降低了大鼠血清中过氧化氢酶、超氧化物歧化酶活性和总抗氧化能力及肝脏中过氧化氢酶、谷胱甘肽过氧化物酶活性和总抗氧化能力(P<0.05),显著提升了大鼠血清中丙二醛、脂质过氧化物和蛋白质羰基含量及肝脏中丙二醛含量(P<0.05);灌服BPP降低了氧化鱼油攻毒大鼠的腹泻率,显著缓解了氧化鱼油攻毒对大鼠血清和肝脏中肝脏功能和抗氧化相关指标以及肝脏中细胞凋亡和抗氧化相关基因表达的负面影响(P<0.05)。综上所述,给大鼠灌服BPP可以缓解氧化鱼油攻毒诱导的腹泻和肝脏损伤,这可能与其通过调节核因子E2相关因子2(Nrf2)信号通路改善抗氧化功能及细胞凋亡有关。The aim of this experiment was to investigate whether the oral administration of Broussonetia papyrifera leaf polysaccharides(BPP)could alleviate the negative effects of the diet containing oxidized fish oil on liver health in rats,and to primarily explore the possible mechanisms.Thirty-two male Wistar rats weaned at 21 days of age(approximate average weight was 58.05 g)were used in a 2×2 factorial design.The trial factors included oral administration of BPP(oral administration of BPP solution or phosphate buffered solution)and oxidized fish oil challenge(feeding the diets containing oxidized fish oil or normal fish oil).The daily oral dose of BPP was 22.5 mg/kg BW.From day 9 to day 26,the challenge of oxidized fish oil was executed.The trial duration was 26 days.The results showed that oxidized fish oil challenge increased diarrhea rate,significantly enhanced glutathione transaminase and glutamic acid transaminase activities in liver and serum and urea nitrogen content in serum of rats(P<0.05),significantly upregulated the mRNA relative expression levels of cysteine aspartate protease 3 and Kelch-like epichlorohydrin-associated protein-1 and the ratio of B lymphocytoma-2-associated X protein to B lymphocytoma-2 in liver of rats(P<0.05),significantly down-regulated the mRNA relative expression levels of B lymphocytoma-2 in liver of rats(P<0.05),significantly decreased catalase and superoxide dismutase activities and total antioxidant capacity in serum of rats(P<0.05),significantly reduced catalase and glutathione peroxidase activities and total antioxidant capacity in liver of rats(P<0.05),significantly increased malondialdehyde,lipid peroxides and protein carbonyls contents in serum of rats(P<0.05),and significantly enhanced malondialdehyde content in liver of rats(P<0.05).Oral administration of BPP reduced diarrhea rate of rats fed the diet containing oxidized fish oil,and significantly alleviated the negative effects of oxidized fish oil challenge on liver-function-associated indices and antioxidant-related i
