摘  要:目的 建立UHPLC Q-Exactive HRMS法鉴定三七极细粉中化合物及其口服吸收后血浆与尿液中的成分。方法 采用Waters BEH C_(18)色谱柱(100 mm×2.1 mm,1.7μm)分析三七极细粉醇提物及口服给药后血浆与尿液的成分;流动相为0.1%甲酸水(A)-乙腈(B),梯度洗脱;流速0.3 ml/min;柱温35℃;电喷雾离子源(ESI),负离子模式。结果 在三七极细粉醇提物中共鉴定出41个化合物,其中38个为皂苷类成分,另3个为黄酮苷类物质。健康男性单剂量(2 g)给予三七极细粉后,血浆及尿液样本中均未检出相应的原型化合物与代谢产物。结论 该法快速、准确,可为三七极细粉质量控制及临床开发提供科学依据。Objective To identify chemical constituents and components in plasma and urine after oral absorption of superfine powder of Notoginseng Radix et Rhizoma by UHPLC Q-Exactive HRMS.Methods A Waters BEH C18 column(100 mm×2.1 mm,1.7μm)was used for gradient elution,using 0.1%formic acid aqueous solution(A)-acetonitrile(B)as mobile phase of gradient elution.The flow rate was 0.3 ml/min,the column temperature was 40 oC,and the electrospray ionization source operated in negative mode was adopted for data acquisition.Results A total of 41 compounds in ethanol extract of the superfine powder of Notoginseng Radix et Rhizoma were identified,among which 38 chemical constituents were saponins and the other 3 were flavonoid glycosides.After oral administration of the superfine powder of Notoginseng Radix et Rhizoma(2 g),no prototype compounds or metabolites were detected in in plasma and urine samples of healthy male subjects.Conclusion The UHPLC Q-Exactive HRMS method is rapid and accurate,and can provide scientific basis for quality control and clinical development of superfine powder of Notoginseng Radix et Rhizoma.
