摘  要:目的:观察黄芩苷对干酵母致热大鼠的解热作用并探讨其作用机制。方法:采用背部皮下注射干酵母构建大鼠发热模型,SD雄性大鼠随机分为正常对照,模型组,阳性组(阿司匹林,0.1 g/kg),黄芩苷高、中、低剂量组(160、80、40 mg/kg),连续给药3 d,测定各组大鼠肛温的变化;酶联免疫法(ELISA)检测血清肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白细胞介素-6(IL-6)、前列腺素E_(2)(PGE_(2))与环磷酸腺苷(cAMP)水平;Western Blot检测各组大鼠脑组织NF-κB p65(核转录因子-κB p65)蛋白表达。结果:黄芩苷高剂量组有显著解热效果(P<0.01),黄芩苷各剂量组均可不同程度降低大鼠血清TNF-α、IL-1β、IL-6、PGE 2和cAMP含量;与正常组比较,模型组脑组织NF-κB p65蛋白表达增多,黄芩苷高剂量组可明显降低大鼠脑组织NF-κB p65表达(P<0.05)。结论:黄芩苷可显著性降低干酵母引起的体温升高,解热机制可能与抑制TNF-α、IL-1β、IL-6、PGE_(2)与cAMP的分泌和减少脑组织NF-κB p65蛋白表达有关。Objective:To study the antipyretic effect of Baicalin on dry yeast induced fever in rats and explore its mechanism.Methods:The rat fever model was established by subcutaneous injection of dry yeast on the back.Male SD rats were randomly divided into normal control group,model group,positive group(Aspirin,0.1 g/kg),Baicalin high-dose,medium-dose and low-dose groups(160,80,40 mg/kg)for 3 days consecutively.The changes of anal temperature in each group rats were determined after modeling.Serum TNF-α,IL-1β,IL-6,PGE_(2) and cAMP were measured by ELISA.Western Blot was used to detect the expression of NF-κB p65 protein in the brain of each group.Results:Baicalin high-dose group had a significant antipyretic effect(P<0.01),and all Baicalin groups reduced the contents of TNF-α,IL-1β,IL-6,PGE_(2) and cAMP in serum of rats to varying degrees.Compared with normal group,the expression of NF-κB p65protein in brain tissue of model group was increased,and the expression of NF-κB p65 in brain tissue of in Baicalin high-dose group was significantly decreased(P<0.05).Conclusions:Baicalin could significantly reduce the temperature increase caused by dry yeast.The antipyretic mechanism may be related to reducing the serum TNF-α,IL-1β,IL-6,PGE_(2),cAMP levels and cerebral NF-κB p65 protein expression.
