摘 要:目的探究司坦唑醇对生长激素缺乏症小鼠氨基酸类神经递质及内分泌生长激素(growth hormone,GH)/胰岛素样生长因子1(insulin like growth factor,IGF)-1的影响。方法随机选择54只小鼠,分为健康组(A组)、模型组(B组)、低剂量司坦唑醇组(C组)、中剂量司坦唑醇组(D组)、高剂量司坦唑醇组(E组)及醋酸泼尼松组(F组),每组9只。给药期间计算小鼠体质量增量,酶联免疫吸附测定血清中氨基酸类神经递质谷氨酸(glutamic acid,Glu)、γ-氨基丁酸(γ-aminobutyric acid,GABA)及GH水平,苏木精-伊红染色法染色观察胫骨组织学形态,免疫印迹法和聚合酶链式反应分别检测海马组织中IGF-1及GH表达。结果与A组相比,B组大鼠体质量、血清中GABA、GH水平、海马组织中IGF-1、GH信使RNA及蛋白表达水平均降低,血清中Glu水平升高(P<0.01);与B组相比,C组大鼠体质量、血清中GABA、GH、海马组织中IGF-1、GH表达均升高(P<0.05),血清中Glu水平降低(P<0.01);与C组相比,D组大鼠体质量、血清中GABA、GH、海马组织中IGF-1、GH表达均升高(P<0.01),血清中Glu水平降低(P<0.01);与D组相比,E组大鼠体质量、血清中GABA、GH、海马组织中IGF-1、GH表达均升高(P<0.01),血清中Glu水平降低(P<0.01),E组与F组大鼠体质量、血清中GABA、Glu、GH、海马组织中IGF-1、GH表达无差异(P>0.05)。结论司坦唑醇可调节GHD小鼠氨基酸类神经递质,下调Glu水平,升高GABA水平,促进内分泌GH/IGF-1信号通路的表达,为GHD治疗提供新的思路。Objective To investigate the effects of Stanozolol on amino acid neurotransmitters and growth hormone(GH)/insulin-like growth factor-1(IGF-1)in growth hormone deficiency mice.Methods Fifty-four mice were randomly divided into healthy group(group A),model group(group B),low-dose Stanozolol group(group C),medium-dose Stanozolol group(group D),high-dose Stanozolol group(group E),and prednisone acetate group(group F),with 9 mice in each group.During the administration period,body weight gain of mice was calculated.The levels of amino acid neurotransmitters glutamic acid(Glu)andγ-aminobutyric acid(GABA)in serum,as well as GH levels were determined by enzyme-linked immunosorbent assay.Hematoxylin-eosin staining was used to observe the histomorphology of tibia.Immunoblotting and polymerase chain reaction were used to detect the expressions of IGF-1 and GH in hippocampus tissue.Results Compared with group A,rats in group B had decreased body weight,serum GABA and GH levels,and IGF-1,GH mRNA,and protein expression levels in hippocampus tissue,while serum Glu level was increased(P<0.01).Compared with group B,rats in group C had increased body weight,serum GABA and GH levels,and IGF-1 and GH expressions in hippocampus tissue,while serum Glu level was decreased(P<0.01).Compared with group C,rats in group D had increased body weight,serum GABA and GH levels,and IGF-1 and GH expressions in hippocampus tissue,while serum Glu level was decreased(P<0.01).Compared with group D,rats in group E had increased body weight,serum GABA and GH levels,and IGF-1 and GH expressions in hippocampus tissue,while serum Glu level was decreased(P<0.01).There were no differences in body weight,serum GABA,Glu,GH,and IGF-1 and GH expressions in hippocampus tissue between group E and group F(P>0.05).Conclusion Stazolol can regulate amino acid neurotransmitters in GHD mice,down-regulate Glu content,increase GABA content and promote the expression of endocrine GH/IGF-1 signaling pathway,which provides a new idea for GHD treatment.
