摘 要:目的观察天龙竭对博来霉素诱导的肺间质纤维化(PF)大鼠肺组织病理学改变,肺指数、肺组织中羟脯氨酸(Hyp)及I型胶原、Ⅲ型胶原含量的影响,探讨其干预PF的作用机制。方法采用气管内滴注博来霉素的方法复制PF大鼠模型。将90只SPF级雄性Wistar大鼠随机分为9组:空白对照组,模型早、晚期组,天龙竭早期中剂量干预组,天龙竭晚期低、中、高剂量干预组,吡非尼酮早、晚期干预组。造模后第7天及第14天起,每天同一时间不同组别灌胃相应药物,连续用药。观察大鼠的生存状态,造模后第28天取材。计算肺指数,经HE、Masson染色观察肺组织病理变化,采用碱水解法测定Hyp含量,Western blot法和荧光定量PCR技术检测大鼠肺组织中I型胶原(COL-I)、Ⅲ型胶原(COL-Ⅲ)蛋白和mRNA表达量。结果天龙竭显著减轻PF大鼠早期肺部炎性细胞浸润,恢复肺泡结构,降低大鼠肺系数,减少肺组织中Hyp含量,降低COL-I、COL-Ⅲ蛋白和mRNA表达,优于模型组(P<0.05)。结论天龙竭能够减少肺组织胶原沉积,降低肺指数,有效改善实验动物肺纤维化,早期干预尤佳。其作用机制可能与抑制COL-I、COL-Ⅲ表达有关。Objective To observe the effects of Tianlongjie(Draconis Sanguis)dried up on the lung coefficients,hydroxyproline(Hyp)and collagen type I and typeⅢcontents in lung tissues of rats with bleomycin-induced pulmonary fibrosis(PF),and to explore its mechanism of action in intervening PF.Methods The PF rat model was replicated by intratracheal drip bleomycin injection.Ninety SPF-grade male Wistar rats were randomly divided into nine groups:blank control group,early and late model group,early and medium dose intervention group of Tianlongjie(Draconis Sanguis),late low,medium and high dose intervention group of Tianlongjie(Draconis Sanguis),and early and late intervention group of pirfenidone.On the 7th and 14th day after modeling,the rats were gavaged with the corresponding drugs at the same time every day in different groups,and the drugs were administered continuously.The survival status of the rats was observed,and the rats were taken on the 28th day after modeling.The lung coefficients were calculated,and the histopathological changes were observed by HE and Masson staining.The Hyp content was determined by the alkaline hydrolysis method,and the amount of COL-I and COL-Ⅲproteins and mRNA phenotypes in the lung tissues of rats were detected by Western blot and fluorescence quantitative PCR.Results Tianlongjie(Draconis Sanguis)dried significantly reduced inflammatory cell infiltration in the early lung of PF rats,restored alveolar structure,reduced lung coefficient,decreased Hyp content in lung tissue,and decreased COL-I,COL-Ⅲprotein and mRNA expression,which was better than the model group(P<0.05).Conclusion Tianlongjie(Draconis Sanguis)dried up can reduce lung tissue gum deposition,decrease lung coefficient and effectively improve pulmonary fibrosis in experimental animals,especially in early intervention.The mechanism of action may be related to the inhibition of COL-I and COL-Ⅲexpression.
