摘  要:目的在前期研究基础上,进一步对比不同电流强度、不同电极连接方式的电针干预对溃疡性结肠炎(ulcerative colitis,UC)模型小鼠的不同疗效,进而筛选电针治疗UC小鼠的最佳刺激参数。方法5%葡聚糖硫酸钠(dextran sodium sulfate,DSS)溶液自由饮用诱导形成小鼠UC模型,选取单侧“天枢”穴、“上巨虚”穴或双侧“上巨虚”穴,以不同电流强度进行电针干预。治疗组分别为电针1组(单侧“天枢”穴、“上巨虚”穴接电针,疏密波,2/100 Hz,0.3 mA)、电针2组(单侧“天枢”穴、“上巨虚”穴接电针,疏密波,2/100 Hz,0.9 mA)、电针3组(双侧“上巨虚”穴接电针,疏密波,2/100 Hz,1.5 mA)、电针4组(双侧“上巨虚”穴接电针,疏密波,2/100 Hz,0.9 mA)。实验第7~12天对小鼠实施干预,每次干预15 min。空白组与模型组不治疗。比较各组小鼠体质量、疾病活动指数(disease activity index,DAI),采用苏木精-伊红(hematoxylin-eosin,HE)染色法观察结肠组织形态,免疫组织化学(immunohistochemistry,IHC)染色法检测各组小鼠结肠组织内髓过氧化物酶(myeloperoxidase,MPO)、中性粒细胞弹性蛋白酶(neutrophil elastase,NE)表达量。结果电针4组DAI值降低,与模型组相比具有显著性差异(P<0.05),结肠组织炎性浸润较其他组减轻明显。治疗组电针1组、电针2组、电针3组、电针4组结肠长度均较模型组增加(P<0.05),结肠组织中MPO、NE表达下降。双侧“上巨虚”穴接电针的疗效优于单侧“天枢”穴、“上巨虚”穴接电针,且双侧“上巨虚”穴接电针电流强度为0.9 mA的疗效优于1.5 mA。结论电针治疗UC小鼠的最佳刺激参数为双侧“上巨虚”穴接电针,疏密波,2/100 Hz,0.9 mA。Objective On the basis of previous studies,the different effects of electroacupuncture intervention with different current intensities and different electrode connection methods on ulcerative colitis(UC)model mice were further compared,and the optimal stimulation parameters for electroacupuncture treatment of UC mice were screened.Methods The UC model of mice was induced by free drinking of 5%dextran sodium sulfate(DSS)solution.Unilateral“Tianshu(ST25)”“Shangjuxu(ST37)”or bilateral“Shangjuxu(ST37)”were selected for electroacupuncture intervention with different current intensities.The treatment groups were electroacupuncture group 1[unilateral“Tianshu(ST25)”and“Shangjuxu(ST37)”acupoints were connected with electroacupuncture,dilatational wave,2/100 Hz,0.3 mA],electroacupuncture group 2[unilateral“Tianshu(ST25)”and“Shangjuxu(ST37)”acupoints were connected with electroacupuncture,dilatational wave,2/100 Hz,0.9 mA],electroacupuncture group 3[bilateral“Shangjuxu(ST37)”acupoints were connected with electroacupuncture,dilatational wave,2/100 Hz,1.5 mA],electroacupuncture group 4[bilateral“Shangjuxu(ST37)”acupoints were connected with electroacupuncture,dilatational wave,2/100 Hz,0.9 mA].On days 7 to 12 of the experiment,the mice were intervened for 15 min each time.Blank group and model group were not treated.The body weight and disease activity index(DAI)of mice in each group were compared.The morphology of colon tissue was observed by hematoxylin-eosin(HE)staining.The expression of myeloperoxidase(MPO)and neutrophil elastase(NE)in colon tissue of mice in each group was detected by immunohistochemistry(IHC)staining.Results The DAI value was reduced in the electroacupuncture group 4,which was significantly different compared with the model group(P<0.05),and the inflammatory infiltration of colonic tissue was reduced significantly compared with the other groups.The colon length of electroacupuncture group 1,electroacupuncture group 2,electroacupuncture group 3 and electroacupuncture g
