摘 要:须癣毛癣菌(Trichophyton mentagrophytes)又称石膏样毛癣菌,对家兔(Oryctolagus cuniculus)具有严重危害性。过氧化物酶体生物合成因子10(peroxisomal biogenesis factor 10,PEX10)与植物病原真菌的药物敏感性具有密切关系,但是其影响动物病原真菌如须癣毛癣菌生长及致病性的机制尚不明晰。本研究首先构建PEX10基因敲除载体,通过根癌农杆菌介导的遗传转化(Agrobacterium tumefaciensmediated transformation,AtMT)和转化子筛选,获得须癣毛癣菌PEX10基因敲除株;生长特性等表型分析显示,PEX10基因敲除株的生长速率显著减慢(P<0.01),对盐酸小檗碱的敏感性显著提高(P<0.001),侵染兔皮肤组织的能力减弱,致病性降低。本研究结果为真菌PEX10基因功能的研究和兔须癣毛癣菌病的防治提供参考。Trichophyton mentagrophytes,also known as T.gypsioides,is severely hazardousness to rabbit(Oryctolagus cuniculus).Peroxisomal biogenesis factor 10(PEX10)gene is closely related to the drug susceptibility of plant pathogenic fungi.However,the mechanism of whether PEX10 gene affects the growth and pathogenicity of animal pathogenic fungi such as T.mentagrophytes is still unclear.This study first constructed a PEX10 gene knockout vector and obtained a PEX10 gene knockout strain of T.mentagrophytes through Agrobacterium tumefaciens-mediated transformation(AtMT)and transformant screening.Phenotypic analysis such as growth characteristics showed that the growth rate of PEX10 gene knockout strains was significantly slowed down(P<0.01),and their sensitivity to berberine hydrochloride was significantly increased(P<0.001).The virulence and pathogenicity of the PEX10 knockout strain to rabbit skin was reduced.The results of this study provide reference for the study of PEX10 gene function and the prevention and treatment of T.mentagrophytes of rabbits.
