摘 要:胱硫醚β-合成酶(cystathionine β-synthase,CBS)通过与胱硫醚γ-裂解酶(cystathionine γ-lyase,CSE)共同作用生成内源性硫化氢(H2S),参与硫氨基酸代谢来调控细胞的生长、增殖、凋亡等正常生理功能。为了解CBS和CSE在成年牦牛子宫中的作用,分别采集妊娠期、黄体期、卵泡期牦牛子宫组织样品,通过qRT-PCR、Western-blot和免疫组织化学等方法检测CBS和CSE基因及其蛋白的相对表达情况。结果显示,CBS mRNA在黄体期牦牛子宫组织中表达量显著高于妊娠期和卵泡期(P<0.05),且妊娠期的表达量显著高于卵泡期(P<0.05);CSE mRNA在妊娠期的表达量最高(P<0.05),黄体期和卵泡期的表达差异性不显著(P>0.05)。CBS蛋白在卵泡期的表达量最高(P<0.05),黄体期和妊娠期的表达差异量不显著(P>0.05);CSE蛋白在卵泡期牦牛子宫组织中表达量显著高于妊娠期和黄体期(P<0.05),且黄体期的表达量显著高于妊娠期(P<0.05)。免疫组织化学定位显示,牦牛子宫内膜组织中有硫化氢合成酶CBS和CSE的表达,并且CBS和CSE在子宫内膜上皮细胞、腺上皮细胞和子宫内膜基质细胞上表达均为阳性。上述结果表明,CBS和CSE在成年牦牛子宫组织中均有表达,且在不同生殖周期CBS和CSE蛋白及其mRNA在牦牛子宫组织中的表达量有明显的变化。Cystathionineβ-synthase(CBS)regulates cell growth,proliferation,apoptosis and other normal physiological functions by interacting with cystathionine y-lyase(CSE)to generate endogenous hydrogen sulfide(HS)for sulfur amino acid metabolism.To understand the roles of CBS and CSE in adult yak uterus,yak uterine tissues were collected during gestation,luteal phase and follicular phase respec-tively,and the relative expression of CBS and CSE genes and their proteins were detected by qRT-PCR,Western-blot and immunohistochemistry.The results showed that the expression of CBS mRNA in yak uterine tissues during luteal phase was significantly higher than that during gestation and follicular phase(P<0.05),and the expression during gestation was significantly higher than that during follicular phase(P<0.05).The expression of CSE mRNA was highest during gestation(P<0.05),and the differences in the expression between luteal and follicular phases were not significant(P>0.05).The expression of CBS protein was highest during follicular phase(P<0.05),and the expression of CBS protein was highest during follicular phase(P>0.05).(P<0.05),and the difference in expression between luteal and gestational stages was not significant(P>0.05);CSE protein expression in yak uterine tissues during the follicular stage was significantly higher than that during the gestational stage and the luteal stage(P<0.05),and the expression during the luteal stage was significantly higher than that during the gestational stage(P<0.05).Immunohistochemical localization showed the expression of hydrogen sulfide synthase CBS and CSE in yak endometrial tissues,and the expression of CBS and CSE was positive on endometrial epithelial cells,glandular epithelial cells and endometrial stromal cells.The expression of CBS and CSE in the uterine tissues of adult yaks was positive in endometrial epithelial cells,glandular epithelial cells and endometrial stromal cells.
