摘 要:【目的】揭示卷丹百合感染根腐病后根际土壤真菌群落结构、多样性变化,为进一步探明根腐病的微生态发病机理和生防菌的筛选及应用提供理论依据。【方法】分别对连作多年地块上患根腐病和健康卷丹百合根际土壤中真菌的ITS1基因进行扩增,并采用高通量测序技术进行测序,分析健康与患病百合根际土壤真菌物种组成及多样性。【结果】α多样性结果显示,卷丹百合健康与患根腐病根际土壤中的真菌群落丰富度、均匀度和多样性无显著差异。PCA分析结果显示,患根腐病百合与健康百合根际真菌群落物种组成存在差异。患根腐病与健康百合根际真菌优势菌门为子囊菌门;散囊菌纲、银耳纲、刺盾炱目、银耳目、小疱毛壳科、丛赤壳科、外瓶霉属等在患病百合根际中相对丰度显著高于健康百合。LEfSe分析结果显示,子囊菌门与Ilyonectria分别在患病与健康百合根际土壤中差异最大;Arcopilus、角菌根菌属、Ascobolus、Paraphaeosphaeria等14类菌群为健康百合根际的标志物种。【结论】连作多年地块健康百合与患根腐病百合根际真菌群落结构有明显差异。卷丹百合根腐病的发生可能与根际土壤中真菌物种结构的改变,尤其是散囊菌纲、外瓶霉属等有害真菌的增殖密切相关。卷丹百合根际土壤中存在大量潜在益生真菌,是进一步筛选根腐病生防菌的潜在资源。【Objective】The change of rhizosphere fungal community structure and diversity of Lilium lancifolium after infection with root rot were revealed,which was expected to provide a theoretical basis for further exploring the microecological pathogenesis and the screening and application of biocontrol fungi for root rot.【Method】The ITS1 region of fungal rRNA gene in rhizosphere soil was amplified and then se⁃quenced by Illumina Miseq high⁃throughput sequencing technology.The sequencing results were used to analyze the fungal structure and di⁃versity.【Result】The alpha⁃diversity analysis showed that there was no significant difference in richness,evenness and diversity in rhizo⁃sphere soil of healthy and root rot plant.The PCA analysis showed that the fungal structure of L.lancifolium with root rot disease was differ⁃ent from that of healthy plant.The dominant phyla in rhizosphere soil of healthy L.lancifolium and root rot L.lancifolium was Ascomycota.Compared with healthy L.lancifolium plants,the relative abundance of Eurotiomycetes,Tremellomycetes,Chaetothyriales,Tremellales,Herpotrichiellaceae,Nectriaceae and Exophiala were significantly higher.According to the LEfSe analysis,Ascomycota and Ilyonectria showed the greatest difference in the rhizosphere soil of diseased and healthy plant,respectively.And 14 types of flora including Arcopilus,Ceratobasidium,Ascobolus,Paraphaeosphaeria and so on were biomarkers in rhizosphere soil of healthy L.lancifolium.【Conclusion】There was significant difference in fungi community composition of rhizosphere soil from healthy and root rot L.lancifolium in the continuous crop⁃ping plots for many years.Occurrence of L.lancifolium root rot was closed to the variation of fungi community structure in rhizosphere soil,especially proliferation of harmful fungi such as Eurotiomycetes and Exophiala.There was a large number of potential probiotics in the rhizo⁃sphere soil of L.lancifolium,which would be a potential resource for further screening of biocontrol micr
