摘  要:【目的】旨在探究牛至香酚是否能有效改善脂多糖(LPS)免疫应激小鼠免疫功能的损伤。【方法】将80只小鼠随机分成空白对照组、模型组和牛至香酚低、中、高剂量组。牛至香酚低、中、高剂量组分别按25、50、100 mg·kg^(-1)的剂量灌胃牛至香酚,空白对照组和模型组按10 mL·kg^(-1)的剂量灌胃橄榄油,每日1次,连续14 d。第15天时,牛至香酚组和模型组小鼠腹腔注射8 mg·kg^(-1)LPS,空白对照组注射等量生理盐水,6 h后采集血液和肠道组织,检测小鼠血清细胞因子(IL-1β、IL-6、TNF-α、IL-10)含量和肠道细胞因子(IL-1β、IL-6、TNF-α、IL-10)、紧密连接蛋白(ZO-1、Occludin、Claudin-1)mRNA的表达水平。【结果】(1)与空白对照组相比,模型组小鼠血清TNF-α、IL-10、IL-1β含量上升(P>0.05),IL-6含量极显著上升(P<0.01);模型组回肠和结肠组织TNF-α、IL-6、IL-1βmRNA表达水平极显著上调(P<0.01),回肠和结肠组织ZO-1 mRNA表达水平下调(P>0.05),结肠组织Occludin、Claudin-1 mRNA表达水平极显著下调(P<0.01)。(2)与模型组相比,牛至香酚试验组小鼠血清IL-6含量极显著下降(P<0.01),IL-1β含量显著下降(P<0.05);牛至香酚高剂量组血清TNF-α含量极显著下降(P<0.01);牛至香酚中、高剂量组血清IL-10含量显著上升(P<0.05)。(3)同模型组相比,牛至香酚中、低剂量组回肠组织IL-1β、TNF-α、IL-6 mRNA表达水平极显著下调(P<0.01);牛至香酚中、高剂量组回肠组织ZO-1、Occludin mRNA表达水平极显著上调(P<0.01);牛至香酚高剂量组结肠组织IL-10 mRNA表达水平极显著上调(P<0.01),TNF-αmRNA表达水平极显著下调(P<0.01),Occludin、Claudin-1 mRNA表达水平极显著上调(P<0.01)。【结论】牛至香酚通过改变小鼠血清细胞因子含量和肠道细胞因子、紧密连接蛋白mRNA的表达水平,调节LPS免疫应激小鼠的免疫功能。【Objective】The study aimed to explore whether oregano can effectively improve the immune function of mice under immune stress caused by lipopolysaccharide(LPS).【Method】Eighty mice were randomly divided into blank control group,model group,and test groups.The test groups were administrated with low(25 mg·kg^(-1)),medium(50 mg·kg^(-1))or high(100 mg·kg^(-1))dose of oregano phenol while the blank control group and model group were administrated with 10 mL·kg^(-1)olive oil once a day for 14 d.At day 15,the test groups and model group were intraperitoneally injected with 8 mg·kg^(-1)LPS,and the blank group were injected with same amount of saline.Then blood and intestinal tissues of mice were collected 6 h later to determine the contents of serum cytokines(IL-1β,IL-6,TNF-αand IL-10)and mRNA expression levels of intestinal cytokines(IL-1β,IL-6,TNF-αand IL-10)and tight junction proteins(ZO-1,Occludin and Claudin-1).【Result】(1)Compared with the blank control group,serum TNF-α(P<0.05),IL-10(P<0.05)and IL-1β(P<0.05)and IL-6(P<0.01)in the model group were significantly increased.The mRNA expression levels of TNF-α,IL-6 and IL-1βin the ileum and colon of the model group were significantly up-regulated(P<0.01),while that of ZO-1 were down-regulated(P<0.05);the mRNA expression levels of Occludin and Claudin-1 in the colon of the model group were down-regulated(P<0.01).(2)Compared with the model group,serum IL-6 and IL-1βwere extremely significantly(P<0.01)and significantly(P<0.05)decreased in all the test groups,serum TNF-αwas only significantly decreased in the high-dose test group(P<0.01),and serum IL-10 was significantly increased(P<0.05)in both medium-and high-dose test groups.(3)Compared with the model group,the mRNA expression levels of IL-1β,TNF-αand IL-6 in the ileum of medium-and low-dose test groups were extremely significantly down-regulated(P<0.01),while those of ZO-1 and Occludin in the ileum of medium-and high-dose groups were up-regulated(P<0.01).Significantly increased mRNA expressi
