摘 要:为探究齐墩果酸(Oleanolic acid,OA)对细菌性腹泻仔猪肠道损伤的保护作用,利用产肠毒素大肠杆菌(Enterotoxigenic Escherichia coli,ETEC)感染断奶仔猪建立肠道损伤模型。将24头35日龄9~11 kg杜×长×大三元杂交仔猪随机分为3组,每组8次重复。试验期为14 d,其中,试验期间对照与ETEC组饲喂基础日粮,ETEC+OA组饲喂基础日粮+0.01%OA。结果表明,饲喂OA预防ETEC诱导的仔猪腹泻,HE染色和ELISA检测表明OA缓解ETEC感染导致的结肠黏膜形态完整,并通过提高紧密连接蛋白的mRNA表达保护结肠屏障功能。在细胞分子层面上,通过透射电镜、免疫荧光观察结肠上皮细胞凋亡情况,实时荧光定量PCR和Western blot检测发现,ETEC诱导仔猪结肠上皮细胞坏死性凋亡,而OA通过下调BAX和上调BCL-2基因和蛋白表达方式抑制ETEC诱导的肠上皮细胞凋亡。以上结果证实OA通过抑制肠道上皮细胞凋亡改善ETEC感染引起的仔猪肠道屏障完整性损伤。To investigate the protective effect of oleanolic acid(OA)on bacterial diarrhea-induced intestinal damage in piglets,a piglet intestinal injury model was established using Enterotoxigenic Escherichia coli(ETEC)infection in weaned piglets.A total of twenty-four 35-day-old 9-11 kg Du x Chang x Da ternary crossbred piglets were randomly divided into three groups with eight replicates in each group.The experimental period was 14 days,in which the control and ETEC groups were fed a basal diet,and the ETEC+OA group was fed a basal diet+0.01%OA during the experimental period.The results showed that feeding OA prevented ETEC-induced diarrhea in piglets,and HE staining and ELISA assay demonstrated that OA mitigated the intact morphology of the colonic mucosa due to ETEC infection and protected the colonic barrier function by increasing tight junction protein mRNA expression protected the colonic barrier function.At the cellular molecular level,apoptosis of colonic epithelial cells was observed using transmission electron microscopy and immunofluorescence,and real-time fluo rescence quantitative PCR and Western blot assays revealed that ETEC induced necrotic apoptosis of colonic epithelial cells in piglets,whereas OA inhibited ETEC-induced apoptosis of intestinal epithelial cells by down-regulating BAX and upregulating BCL-2 expression.These results confirmed that OA could ameliorate ETEC infection-induced impairment of intestinal barrier integrity in piglets by inhibiting intestinal epithelial cell apoptosis.
