摘  要:克隆与人参皂苷Rg_(2)合成相关的重要酶基因PgRg_(2)BBE全长cDNA序列,并利用在线软件对该基因编码的蛋白质进行理化性质、结构域、亚细胞定位和系统进化等分析。结果表明:PgRg_(2)BBE基因cDNA序列长为1638 bp,编码545个氨基酸。PgRg_(2)BBE蛋白分子量为60967.10 u,等电点为8.88,不稳定性指数为34.04,预测该蛋白为一种稳定的亲水性跨膜蛋白,主要由α-螺旋和无规则卷曲组成。PgRg_(2)BBE蛋白含有FAD_binding_4和BBE功能结构域,定位于叶绿体中,含有24个氨基酸序列信号肽。系统发育树分析发现,其与智利茄的亲缘关系最近,氨基酸序列多重比对发现,RgRg_(2)BBE中含有4个保守基序,进一步与拟南芥AtBBE家族蛋白进行同源序列比对,发现它们同时具有4个保守基序(RKYGL、MGED、FWAIRGG、FPEI)。The syudy cloned the full-length cDNA sequence of PgRg_(2)BBE,an important enzyme gene related to ginsenoside Rg_(2) synthesis,and analyzed the physical and chemical properties,structural domain,subcellular localization and systematic evolution of the gene encoding protein usingh online software.The results showed that the cDNA sequence of PgRg_(2)BBE was 1638 bp in length and encoded 545 amino acids.The molecular weight of PgRg_(2)BBE protein was 60967.10 u,the isoelectric point was 8.88,and the instability index was 34.04.It is predicted that the protein is a stable hydrophilic transmembrane protein,mainly composed ofα-helix and random coil.PgRg_(2)BBE protein contains FAD_binding_4 and BBE functional domain,which is located in chloroplast and contains 24 amino acid sequence signal peptides.Phylogenetic tree analysis showed that it had the closest genetic relationship with Chilean eggplant,and multiple amino acid sequence alignment showed that RgRg_(2)BBE contained four conserved motifs.Further homologous sequence comparison with Arabidopsis AtBBE family proteins showed that there were four conserved motifs(RKYGL,MGED,FWAIRGG and FPEI).
