摘 要:目的 利用代谢组学技术探究痹祺胶囊治疗类风湿性关节炎的作用机制。方法 建立II型胶原蛋白诱导的类风湿性关节炎(rheumatoid arthritis,RA)大鼠模型,并进行痹祺胶囊给药干预,取空白组、模型组、给药组大鼠血清,采用UPLCQ-TOF-MS技术对血清进行代谢组学分析,筛选潜在生物标志物,结合人类代谢组学数据库(human metabolomics database,HMDB)和京都基因与基因组百科全书数据库(Kyoto encyclopedia database of genes and genomes,KEGG)富集相关代谢通路并分析痹祺胶囊对生物标记物的干预作用,进而解析其作用机制。结果 代谢组学分析共筛选到18个潜在生物标志物,涉及苯丙氨酸、酪氨酸和色氨酸的生物合成及苯丙氨酸代谢、醚脂代谢、组氨酸代谢、三羧酸循环、精氨酸生物合成等11条代谢通路;痹祺胶囊可改善模型大鼠代谢轨迹的偏离。结论 痹祺胶囊主要通过调节氨基酸代谢、花生四烯酸代谢、甘油磷脂代谢及能量代谢等多条代谢通路,改善内源性代谢物的水平,在机体供能、抗炎及免疫调节过程中发挥作用。Objective To study the mechanism of Biqi Capsules(BQ,痹祺胶囊)in the treatment of rheumatoid arthritis by metabolomics technology.Methods The rat model of rheumatoid arthritis(RA)induced by type II collagen was established,and the intervention of BQ was carried out.The serum of blank group,model group and administration group was taken,and UPLC-Q-TOFMS technology was used to analyze the metabolomics of serum to screen potential biomarkers.Combined with human metabolomics database(HMDB)and Kyoto Encyclopedia of Genes and Genomes(KEGG),the related metabolic pathways were enriched and the intervention effect of BQ on biomarkers was analyzed to elucidate its mechanism.Results A total of 18 potential biomarkers were screened by metabolomics analysis,involving 11 metabolic pathways such as phenylalanine,tyrosine and tryptophan biosynthesis,phenylalanine metabolism,ether lipid metabolism,histidine metabolism,TCA cycle,arginine biosynthesis.BQ can improve the deviation of metabolic trajectory in model rats.Conclusion BQ mainly improves the level of endogenous metabolites by regulating multiple metabolic pathways such as amino acid metabolism,arachidonic acid metabolism,glycerophospholipid metabolism and energy metabolism,and plays a role in energy supply,anti-inflammation and immune regulation.
