海藻药材中砷元素形态及价态分析研究

摘  要:海藻是一类重要的中药材,具有多种药理活性,但也可能富集海水中的重金属元素,比如砷。砷是一种广泛存在于自然界的有毒元素,其不同的化学形态及价态具有不同的毒性。因此,准确测定海藻中砷的含量和形态及价态对于评估其食用安全和药用价值具有重要意义。本文采用高效液相色谱-电感耦合等离子体质谱联用法(HPLC-ICP-MS),建立了一种同时测定海藻中砷胆碱(AsC)、砷甜菜碱(AsB)、亚砷酸(As(Ⅲ))、二甲基砷(DMA)、一甲基砷(MMA)、砷酸(As(V))的方法。该方法各形态及价态砷线性相关系数均在0.9991以上,检出限(LOD)为0.0002~0.002 mg/kg,相对标准偏差(RSD)为0.48%~3.44%,回收率为80.22%~107.43%。10批海藻药材的检测结果表明,海藻中总砷及有毒的无机砷含量远高于相关药品及食品标准。Sargassum,an important type of Chinese herbal medicine with various pharmacological activities,has the potential to accumulate heavy metal elements,such as arsenic,from seawater.Arsenic,a toxic element widely distributed in nature,exhibits varying toxicities with different chemical forms and valence states.Therefore,accurately measuring the content,forms and valence states of arsenic in sargassum is of great significance for evaluating its edible safety and medicinal value.This paper establishes a method for the simultaneous determination of Arsenic choline(AsC),Arsenobetaine(AsB),Arsenous acid[As(III)],dimethyl arsenic(DMA),monomethyl arsenic(MMA),arsenate[As(V)]in sargassum by high-efficiency liquid chromatography-inductively coupled plasma mass spectrometry(HPLC-ICP-MS).The linear correlation coefficients for each form and valence state of arsenic in this method were above 0.9991,with detection limits(LOD)ranging from 0.0002~0.002 mg/kg,relative standard deviations(RSD)between 0.48%and 3.44%,and recovery rates of 80.22%~107.43%.The test results of 10 batches of sargassum medicinal materials indicated that the total arsenic and toxic inorganic arsenic content in sargassum are significantly higher than t he relevant standards for drugs and food.
