茉莉缓慢生长离体保存技术研究

摘　　要：为了寻找茉莉种质资源缓慢生长离体保存的有效方法,延长继代培养时间,以茉莉组培苗为试验材料,比较植物生长延缓剂ABA及调节渗透物质甘露醇对茉莉组培苗在常温25℃、15℃和10℃下的保存效果。结果表明:在培养基中添加ABA有利于茉莉组培苗的延缓生长,适宜的ABA浓度为1 mg/L;当培养基中添加甘露醇后,所有处理的株高、节数、叶片数均比对照CK低,表明添加甘露醇后也能一定程度上达到延缓生长的目的,筛选出的甘露醇浓度为20 g/L;在添加适宜浓度的ABA与甘露醇后,结合低温环境处理,植株的存活率明显提升,低温10℃的保存效果优于常温处理。To determine an effective method for retarding growth of Jasminum sambac germplasm resources in vitro and extending secondary culture time,this study evaluates the preservation effects of the plant growth inhibitor ABA and permeant mannitol on test tube seedlings at 25℃,15℃and 10℃.The results indicate that the addition of ABA to the medium facilitates the growth retardation of jasmine seedlings,with an effective ABA concentration of 1 mg/L.Similarly,when the culture medium was supplemented with mannitol,the plant height,node,and leaf number in all treatments were lower than those in the CK treatment.This suggests that the addition of mannitol to the medium also facilitates growth retardation,especially with an optimal concentration of 20 g/L.After the addition of suitable concentrations of ABA and mannitol,coupled with low-temperature environment treatment,the plant survival rate significantly increased.The preservation effect at low temperature of 10℃surpassed that observed at room temperature.
