掺钆纳米氧化铈抑制ApoE基因敲除小鼠动脉粥样硬化形成的观察研究

摘  要:目的 观察掺钆纳米氧化铈对抗ApoE基因敲除(ApoE-/-)小鼠动脉粥样硬化形成。方法将20%掺钆Ce-Gd纳米制剂根据不同浓度分为:生理盐水对照组、25μg/ml组、50μg/ml组和100μg/ml组,观察该制剂对脂多糖(LPS)处理后ApoE-/-小鼠血浆炎症因子水平的影响。建立ApoE-/-小鼠动脉粥样硬化模型,20%掺钆Ce-Gd纳米制剂根据不同浓度进行分组:生理盐水输注组、10 mg/kg Ce-Gd输注组、50 mg/kg Ce-Gd输注组(输注时间点为0 d,3 d,7 d),油红O染色法观察各组小鼠主动脉粥样硬化面积并进行统计学分析。结果 掺钆20%Ce-Gd的纳米制剂可浓度依赖性降低LPS诱导的血浆炎症因子水平,掺钆20%Ce-Gd的纳米制剂可浓度依赖性降低Apo E-/-小鼠动脉粥样硬化斑块负荷。结论 掺钆纳米氧化铈通过降低Apo E-/-小鼠氧化应激水平,对抗动脉粥样硬化形成,有望成为新型抗动脉粥样硬化治疗药物。Objective To observe the antagonism of gadolinium doped nano cerium oxide(Ce-Gd) to formation of atherosclerosis(AS) in mice with ApoE gene knockout(ApoE-/-).Methods Ce-Gd(20%) were divided,according to different doses,into normal saline control group(NS group),25 μg/ml group(low-dose group),50 μg/ml group(mid-dose group) and 100 μg/ml group(high-dose group).The influence of Ce-Gd,after treated with lipopolysaccharide(LPS),on levels of plasma inflammatory factors were observed in ApoE-/-mice.A model of ApoE-/-mice with AS was established,and divided into NS infusion group,10 mg/kg Ce-Gd infusion group(lowdose Ce-Gd group) and 50 mg/kg Ce-Gd infusion group(high-dose Ce-Gd group) at time points of 0 d,3 d and 7 d.The area of aortic AS was observed by using oil red O staining,and statistical analysis was carried out in all groups.Results Ce-Gd can decreased levels of plasma inflammatory factors induced by LPS,and AS loading in a doseindependent manner in ApoE-/-mice.Conclusion Ce-Gd can inhibit AS formation through reducing oxidative stress level in ApoE-/-mice,which is expected to become a new anti-atherosclerosis medicinal.
