摘 要:目的:研究鸿茅药酒的活血化瘀作用。方法:采用FeCl 3诱导颈总动脉血栓模型,检测鸿茅药酒对活化部分凝血酶原时间(APTT)、凝血酶原时间(PT)、凝血酶时间(TT)及血栓湿重、6-酮-前列腺素F1α(6-keto-PGF 1α)、血栓素B2(TXB 2)、P-选择素、血管性血友病因子(vWF)、环磷酸腺苷(cAMP)的影响;采用角叉菜胶致小鼠尾部血栓模型、断尾法,观察鸿茅药酒对小鼠血栓形成、出血时间的影响;采用皮下注射盐酸肾上腺素加冰水浴复制的大鼠急性血瘀模型,检测鸿茅药酒对急性血瘀模型大鼠血液流变学、6-keto-PGF 1α、TXB 2的影响。结果:鸿茅药酒能延长动脉血栓模型大鼠APTT、PT、TT时间,减少血栓湿重,减少P-选择素、vWF、TXB 2含量,增加cAMP含量;明显减少小鼠尾部血栓长度;降低急性血瘀模型大鼠全血、血浆黏度和红细胞压积,增加6-keto-PGF 1α含量,减少TXB 2含量;且中、低剂量组无明显延长小鼠的出血时间。结论:鸿茅药酒能显著抑制体内血栓形成,改善凝血系统,调节PGI 2/TXA 2动态平衡,改善血瘀大鼠的血液流变学异常变化,具有一定抗血栓作用,出血风险较小。Objective:To study the effects of Hongmao Medication Wine(HMW)on promoting blood circulation for dissipating blood stasis action.Methods:The common carotid artery thrombosis rat model induced by FeCl 3 were established to detect the effects of HMW on the activated partial thromboplastin time(APTT),prothrombin time(PT)and thrombin time(TT)and thrombus wet weight and 6-keto-prostaglandin F1α(6-keto-PGF 1α),Thromboxane B2(TXB 2),P-selectin,Von Willebrand factor(vWF),cyclic adenosine monophosphate(cAMP)were detected..The mice were treated carrageenan to induced mice tail thrombus model and cutting tail tip method,and the effects of HMW on antithrombotic and bleeding time were determined.Then the rats were treated with adrenaline hydrochloride subcutaneously and ice-water bath to induced acute blood stasis model,and the effects of Hongmao Medication Wine on blood rheological parameters and 6-keto-PGF 1αand TXB 2 were detected.Results:HMW could prolong the time of APTT,PT and TT times,and reduce wet weight of thrombus and the content of P-selectin,vWF,TXB 2,and increase the content of cAMP in the rats with arterial thrombosis model significantly.The length of tail thrombus in mice were significantly reduced by the treatment of HMW.Reduce whole blood viscosity,plasma viscosity and hematocrit were reduced,and 6-keto-PGF 1αcontent was increased,TXB 2 content was reduced in the rats with acute blood stasis model.Besides,HMW middle-dose and low-dose groups had no significant effect on the bleeding time of mice.Conclusion:HMW could obviously have anticoagulant effects and ameliorate anticoagulant system.It also ameliorate cameliorate hemorheological abnormalities and the regulation of PGI 2/TXA 2 homeostasis in acute blood stasis rats.These effects suggest HMW had a certain Antithrombotic effects.
