高效液相色谱法同时测定万古霉素和去甲万古霉素的血药浓度分析

摘　　要：目的:分析高效液相色谱法(HPLC)同时测定血浆中万古霉素和去甲万古霉素血药浓度。方法:采用Phenomenex C 18柱的色谱柱,流动相为0.05 mol·L^(-1) KH 2PO 4:乙腈(90:10);流速;1mL·min^(-1);检测波长为236nm;以万古霉素与去甲万古霉素作为内标物。结果:万古霉素在1.51~89.12μg·mL^(-1)和去甲万古霉素在1.34~84.57μg·mL^(-1)间具有很好的内线性关系(r>0.99);万古霉素的回收率为88.51%~93.70%,去甲万古霉素为90.56%~94.68%,日内、日间RSD在13%以内。结论:HPLC同时测定血浆中万古霉素和去甲万古霉素血药浓度操作简便,结果准确。Objective:Simultaneous determination of vancomycin and norvancomycin concentrations in plasma by high-performance liquid chromatography(HPLC).Method:Using Phenomenex C18 columns A chromatographic column with a mobile phase of 0.05 mol·L^(-1) KH 2PO 4:acetonitrile(90:10);Flow rate;1mL·min-1;The detection wavelength is 236nm;Use vancomycin and norvancomycin as internal standards.Result:The linear relationship between vancomycin and norvancomycin between 1.34~84.57μg·mL^(-1) and 1.51~89.12μg·mL^(-1) was very good(r>0.99).The recoveries of vancomycin and norvancomycin were 88.51%~93.70% and 90.56%~94.68% respectively.The RSD of intra-day and intra-day were within 13%.Conclusion:Simultaneous determination of vancomycin and norvancomycin in plasma by HPLC is simple and accurate.
