雏鸡脂肪肝冰冻切片制作过程中苏丹Ⅲ染色条件的探索

摘　　要：为探讨在雏鸡脂肪肝冰冻切片制作过程中苏丹Ⅲ染色的最佳条件,取7日龄雏鸡的肝脏组织,用冰冻切片机制作成切片,然后设置不同条件,用苏丹Ⅲ染色液对切片进行染色,观察染色效果,进行对比.结果表明:用苏丹Ⅲ染料染色9 min明显好于5 min和30 min,70%乙醇溶液冲洗5次明显好于2次和9次,蓝化3 min比未蓝化效果更佳.将肝组织冰冻切片标本用苏丹Ⅲ浸染9 min、70%乙醇溶液冲洗5次、苏木素浸染1 min、在自来水中蓝化3 min,肝组织的脂肪部位被明显染成橘红色,观察效果最好.In order to explore the optimal conditions for Sudan III staining in the production of frozen sections of chicken fatty liver,this experiment took liver tissue from 7-day-old chickens and made them into sections using a frozen slicer.Then,different conditions were designed to stain the sections with Sudan III staining solution,and the staining effect was observed and compared.The results showed that dyeing with Sudan III dye for 9 min was significantly better than 5 and 30 min,washing with 70%alcohol 5 times was significantly better than 2 and 9 times,and the effect of blueing for 3 min was better than that of not blueing.Conclusion:The frozen section specimens of liver tissue were stained with Sudan III for 9 min,washed 5 times with 70%alcohol,soaked with hematoxylin for 1 minute,and blued in tap water for 3 min.The fatty parts of liver tissue were significantly stained orange red,and the observation effect was the best.
