摘 要:目的研究氮化硅/氧化钇/氧化镁复合陶瓷粉末浸提液对体外培养大鼠胚胎神经细胞毒性的影响。方法利用大鼠胚胎神经细胞进行原代培养,将不同浓度氮化硅/氧化钇/氧化镁复合陶瓷材料粉末浸提液(50、100、200、400μg/mL)分别设置为复合陶瓷实验组1、2、3、4,另设正常对照组、阳性对照组(醋酸铅浓度50μg/mL)。采用神经元特异性烯醇化酶(NSE)免疫组织化学法对神经细胞进行鉴定,并利用图像分析细胞形态的变化。通过四唑盐比色试验(MTT)测定大鼠胚胎神经细胞相对增殖率(RGR)、考马斯亮蓝G-250测定蛋白质相对含量。结果50~400μg/mL氮化硅/氧化钇/氧化镁复合陶瓷浸提液能促进体外培养中脑神经细胞突起生长,集落数增多。阳性染色分布于神经元的胞质及突起中颗粒状。与正常对照组比较,各实验组神经细胞集落数生长分化明显升高、第7天神经细胞相对蛋白质含量明显升高(P<0.01);而阳性对照组(醋酸铅)较各实验组神经细胞集落数生长分化明显抑制、蛋白质含量明显降低(P<0.01)。4个实验组RGR分别为82.3%、90.8%、92.8%、83.6%。细胞毒性为1级,阳性对照组(醋酸铅)细胞RGR为46.9%,细胞毒性为4级。结论氮化硅/氧化钇/氧化镁复合陶瓷粉末浸提液浓度50~400μg/mL为适宜。氮化硅/氧化钇/氧化镁复合陶瓷对体外培养胚胎中脑神经细胞分化、增殖具有一定的促进作用,具有双重性影响0x0E0xFF0xF00xFF倎0x030x0F其生物学作用性质与剂量有关,该材料具有较好的生物相溶性,生物效应是无毒。Objective To study the effect of silicon nitride/yttrium oxide/magnesium oxide composite ceramic powder on the toxicity of rat embryonic nerve cells in vitro.Methods Rat embryonic nerve cells were primarilycultured and silicon nitride/yttrium oxide/magnesium oxide composite ceramics were observed at different concentrations 50,100,200,400μg/mL.Neuron-specific enolase(NSE)immunohistochemistry was used to identify the nerve cells,and the morphological changes of the cells were analyzed by images.The relative growth rate(RGR)of rat embryonic nerve cells was determined by tetrazolium salt colorimetric assay(MTT),and the relative protein content of Coomasil bright blue G-250 was determined.Results 50-400μg/mL silicon nitride/yttrium oxide/magnesium oxide composite ceramic extract promoted the growth of neurite cells in vitro and increased the number of colonies.The positive staining was distributed in granular form in the cytoplasm and processes of neurons.There were significant differences in variance analysis among the 4 concentration composite ceramic groups(P<0.05 or P<0.01).The relative RGRs were 82.3%,90.8%,92.8%and 83.6%,respectively.In the control group(lead acetate),the RGR was 46.9%,and the cytotoxicity was grade 4.The relative protein content of each concentration was statistically significant(P<0.05 or P<0.01).In the control group(Pb acetate)at 50μg/mL,the cell colony formation rate and intercellular nerve fibers weresignificantlydecreased,indicating that Pb acetate could inhibit cell differentiation.Conclusion The concentration of silicon nitride/yttrium oxide/magnesium oxide composite ceramic powder extract is 50-400μg/mL.Silicon nitride/yttrium oxide/magnesium oxide composite ceramics can promote the differentiation and proliferation of embryonic mescranial nerve cells in vitro,and has a dual effect.Its biological properties are dose-dependent,and the material has a good biocompatibility and non-toxic biological effects.
