响应面法优化蝉花多糖热水提取工艺研究

摘  要:为了研究蝉花多糖提取工艺,采用热水提取乙醇沉淀富集粗多糖,Sevag法脱蛋白、活性炭法脱色制备精制多糖计算换算因子以及蝉花多糖提取率,应用单因素实验考察液料比、提取次数、提取温度和提取时间等关键工艺参数对蝉花多糖提取效果的影响,在此基础上采用四因素三水平响应面法优化蝉花多糖提取工艺参数,得到的最优提取条件为:液料比57∶1(mL∶g),提取温度82℃,提取123 min,提取两次。按照此实验条件,所得蝉花多糖提取率为13.88%,与预测值基本相符,说明该工艺稳定、可靠,为蝉花多糖的开发利用提供数据支持。In order to study the extraction process of polysaccharides from cordyceps cicadae(PCC),the crude polysaccharide was enriched by ethanol precipitation from hot water extraction.The refined polysaccharide was prepared by Sevag deproteinization and activated carbon decolorization,which was used to calculate the conversion factor and extraction rate of PCC.The influence of key process parameters such as liquid-solid ratio,extraction times,extraction temperature and extraction time on the extraction effect was investigated by single factor experiment.On this basis,the four-factor and three-level response surface method was adopted to optimize the extraction process parameters of PCC.The optimal extraction conditions were as follows:Liquid-solid ratio 57∶1(mL∶g),extraction temperature 82℃,extraction time 123 min,extraction times 2 times.According to the experimental conditions,the extraction yield of PCC was 13.88%,which was basically consistent with the predicted value,indicating that the process was stable and reliable.To provide data support for the exploitation and utilization of PCC.
