摘  要:miR-140-3p是在不同物种性腺组织中广泛表达的一种miRNA。本研究利用qRT-PCR方法检测miR-140-3p在大鳞副泥鳅性腺中的雌雄表达差异并预测其生物学功能。结果显示:miR-140-3p在大鳞副泥鳅雌雄鱼的性腺组织中均有表达,且精巢中的表达量显著高于卵巢(P<0.01);KEGG pathway分析表明,靶基因CPEB4和GDF9参与卵母细胞成熟分化过程,富集在与性腺发育、繁殖功能相关的孕激素介导卵母细胞成熟通路、卵母细胞减数分裂通路、卵巢类固醇生成通路。荧光定量结果显示CPEB4和GDF9在卵巢中表达量显著高于精巢(P<0.01),且表达量与miR-140-3p的表达呈显著负相关,说明miR-140-3p和CPEB4、GDF9具有潜在的靶标关系。这为进一步探索miR-140-3p在鱼类性腺发育和繁殖过程中的作用奠定了基础。MiR-140-3p is a miRNA widely expressed in gonadal tissues of different species.qRT-PCR was used to detect the expression difference between male and female of miR-140-3p in gonads of Parabgurnus dabryanus,and its biological function was analyzed by predicting related target genes.The results showed that miR-140-3p was expressed in both testis and ovaries,and the expression level of miR-140-3p in testis was significantly higher than that in ovary(P<0.01).KEGG pathway analysis showed that target genes CPEB4 and GDF9 were involved in oocyte maturation and differentiation,and were enriched in progesterone mediated oocyte maturation pathway,oocyte meiosis pathway and ovarian steroid generation pathway,respectively.These pathways were related to gonad development and reproduction.qRT-PCR results showed that the expression of CPEB4 and GDF9 in ovary was significantly higher than that in testis(P<0.01),and the expression level was significantly negatively correlated with expression of miR-140-3p,and these results suggest that miR-140-3p has a potential target relationship with CPEB4 and GDF9.This study provides basic information for future exploration of the role of miR-140-3p in gonad development and reproduction of fish.
