补肾助孕方对LPD模型大鼠子宫内膜NF-κB、NLRP3、COX-2表达水平的影响

摘  要:目的研究补肾助孕方对于由米非司酮诱导的LPD模型大鼠子宫内膜炎性因子NF-κB、NLRP3、COX-2表达水平的影响,探讨补肾助孕方治疗LPD的相关机制。方法随机将60只雌性SD育龄期大鼠分为空白组、模型组、阳性药地屈孕酮组、补肾助孕方低、中、高剂量治疗组,每组10只进行相应造模给药。分别采用qPCR、Western Blot法检测各组大鼠子宫内膜NF-κB、NLRP3、COX-2的mRNA和蛋白表达水平。结果与空白组相比,模型组NF-κB、NLRP3的蛋白及mRNA表达水平明显上升(P<0.01),COX-2蛋白及mRNA表达水平明显下降(P<0.01)。与模型组相比,NF-κB中药各剂量组的蛋白及mRNA表达均下调(P<0.05),但地屈孕酮组无统计学差异(P>0.05);NLRP3阳性药、补肾助孕方低、中、高剂量治疗组的mRNA均明显下降(P<0.05),但蛋白水平,仅中、高剂量中药组有统计学差异;补肾助孕方刺激了COX-2表达,其蛋白表达水平在各组间有显著上升(P<0.05),其中中剂量组效果最佳,mRNA水平仅中药高剂量组有统计学差异。结论补肾助孕方可能通过提高LPD模型大鼠子宫内膜容受性,提高抗氧化、抗炎能力,缓解大鼠子宫内膜组织凋亡、炎性水平,达到治疗LPD目的。Objective To study the effect of Bushen Zhuyun Decoction(BSZYD)on the expression level of endometrial inflamma-tory factors NF-κB,NLRP3 and COX-2 in LPD model induced by mifepristone,and to explore the related mechanism of BSZYD in treating LPD.Methods 60 female SD rats of childbearing age were randomly divided into blank group,model group,positive drug dydrogesterone group,and treatment groups with low,medium,and high doses of BSZYD,with 10 rats in each group were given corresponding modeling and administration.The mRNA and protein expression levels of NF-κB,NLRP3 and COX-2 in each group were respectively by qPCR and Western Blot.Results Compared with the blank group,the expression lev-els of NF-κB,NLRP3 protein and mRNA in the model group were significantly increased(P<0.01),while the expression lev-els of COX-2 protein and mRNA were significantly decreased(P<0.01).Compared with the model group,NF-κB the ex-pression of protein and mRNA in each dose group of BSZYD was down regulated(P<0.05),but there was no statistical differ-ence in the dydrogesterone group(P>0.05);The mRNA levels of NLRP3 positive drugs and low,medium,and high doses of BSZYD groups were significantly decreased,but the protein levels were only statistically different between the medium and high doses of BSZYD;BSZYD stimulated the expression of COX-2,and the protein expression level increased significantly among the groups(P<0.05),the medium dose group showed the best effect,and the mRNA level was only statistically different in the high dose group of BS2YD.Conclusion BSZYD may achieve the goal of treating LPD by improving the endometrial receptivity of LPD model rats,improving the antioxidant and anti-inflammatory capabilities,and alleviating the level of apoptosis and inflammation in rat endometrial tissues.
