摘  要:目的通过毒性成分黄独素B含量差异分析初评绿豆药汁制黄药子的炮制减毒作用及其工艺。方法以绿豆质量分数(A)、闷润时间(B)、炮制温度(C)、炮制时间(D)为因素,采用L9(34)正交试验设计制备绿豆药汁制黄药子各炮制品,以C_(18)反相色谱柱(4.6 mm×250 mm,5μm)、流动相乙腈-水(28∶72)、进样量10μL、流速1.0 mL·min^(-1)、柱温为30℃、检测波长210 nm为色谱条件,通过高效液相色谱法对黄药子生品及其正交试验设计下的各炮制品中黄独素B含量的检测分析,初评绿豆药汁制黄药子的炮制减毒作用及其工艺。结果黄药子生品中毒性成分黄独素B含量为1.813 mg·g^(-1),正交试验设计下绿豆药汁制黄药子各炮制品中毒性成分黄独素B的含量在1.228~1.629 mg·g^(-1),较黄药子生品中显著下降10.18%~32.26%(P<0.01),其中尤以A3B1C3D2工艺下的绿豆药汁制黄药子炮制下降32.26%为最优。结论从毒性成分黄独素B含量差异层面表明,绿豆药汁制黄药子具有炮制减毒作用,A3B1C3D2是其最佳炮制减毒工艺。Objective A preliminary evaluation of the concoction and toxicity reduction effect of Phaseolus radiatus juice made from Dioscorea bulbifera and its process by the analysis of the difference in the content of the toxic component Diosbulbin B.Methods Using P.radiatus mass fraction(A),quenching time(B),processing temperature(C)and processing time(D)as factors,the preparative products from P.radiatus juice to D.bulbifera were prepared by L 9(34)orthogonal design.The chroma-tography was carried out on a C_(18) reverse phase column(4.6 mm×250 mm,5μm)and mobile phase acetonitrile-water(28:72),the sample size was 10μL,the flow rate was 1.0 mL·min^(-1),the column temperature was 30℃,and the detection wave-length was 210 nm as the chromatographic conditions.The content of Diosbulbin B in D.bulbifera and its various products under the orthogonal experimental design were detected and analyzed by HPLC,and the processing effect and technology of D.bulbifera from the medicinal juice of P.radiatus were evaluated.Results The content of toxic ingredient Diosbulbin B in D.bulbifera was 1.813 mg·g^(-1),and the content of toxic ingredient flavin B in the various products of D.bulbifera prepared from P.radiatus juice was 1.228~1.629 mg·g^(-1) under orthogonal design,which was significantly decreased by 10.18%~32.26%compared with D.bulbifera(P<0.01).Among them,the processing of D.bulbifera from P.radiatus juice decreased by 32.26%under A 3 B 1 C 3 D 2 technology.Conclusion From the level of difference of the content of the toxic ingredient Diosbulbin B,the results showed that the D.bulbifera made from P.radiatus juice had the processing attenuating effect,and A 3 B 1 C 3 D 2 was the best pro-cessing attenuating technology.
