摘  要:目的:探讨交感神经释放递质去甲肾上腺素(Norepinephrine,NE)对小鼠心肌细胞增殖的影响。方法:在原代培养的新生小鼠心肌细胞上,采用免疫荧光和Edu荧光标记方法观察NE、β受体激动剂和阻断剂对心肌细胞增殖的影响;在出生后14 d的小鼠上,采用急性酶分离和荧光染色方法观察药物干预β受体对心肌细胞成核的影响。结果:在原代培养的心肌细胞上,NE、β1和β2受体激动剂均能明显降低心肌细胞的Edu阳性率(P<0.05);药物抑制β1受体能够阻断NE对心肌细胞增殖的抑制作用(P<0.05)。在发育期小鼠上,阻断β受体能明显增加单核心肌细胞率,降低多核心肌细胞率(P<0.05)。结论:NE可以通过β1受体对心肌细胞的增殖起到抑制作用。Objective:To investigate the effects of sympathetic neurotransmitter,norepinephrine(NE)on the proliferation of mouse cardiomyocytes.Methods:The effects of NE,βreceptor agonists and blockers on the proliferation of primary cultured neonatal mouse cardiomyocytes were observed by immunofluorescence and Edu fluorescence labeling methods.Acute enzyme isolation and fluorescent staining were used to observe the effect ofβ-AR blocker on cardiomyocyte nucleation in mice 14 days after birth.Results:On primary cultured cardiomyocytes,NE,β1 andβ2 agonists could significantly reduce the Edu positive rate of cardiomyocytes(P<0.05).The chemical inhibition ofβ1 receptor could block the inhibitory effect of NE on cardiomyocyte proliferation(P<0.05).In developing mice,blockingβreceptor significantly increased the rate of mononuclear cardiomyocytes and decreased the rate of multinucleated cardiomyocytes(P<0.05).Conclusion:NE can inhibit the proliferation of cardiomyocytes throughβ1 receptor.
