摘  要:试验探讨冷暴露条件下添加盐酸川芎嗪对小鼠肝脏产热蛋白PGC-1α的影响。试验选取C57BL/6雄性小鼠40只,预饲一周后,随机分为四组。常温对照组保持在26±2℃环境中饲养,冷暴露组和加药组每天置于4℃人工气候室中冷刺激3 h,加药组小鼠在冷暴露前进行相应的灌胃处理,四周后处死小鼠并采集肝脏组织。采用HE染色法观察肝脏组织是否有损伤;采用油红O染色法观察肝脏组织中脂滴数量;采用IHC法和Western Blot法对肝脏组织中PGC-1α表达量进行检测。结果显示,与对照组相比,添加盐酸川芎嗪后肝脏无损伤,脂滴数增加,PGC-1α的表达量显著提高。综上,冷暴露条件下盐酸川芎嗪可提高PGC-1α的表达量,促进小鼠肝脏产热。To investigate the effect of ligustrazine hydrochloride on liver heat producing protein PGC-1αin mice under cold exposure,40 C57BL/6 male mice were randomly divided into four groups after pre-feeding for one week.The normal temperature control group was kept at 26±2℃,and the cold exposure group and the drug addition group were placed in the artificial climate chamber at 4℃for cold stimulation for 3 h every day.The mice in the drug addition group were given corresponding gavage before cold exposure,and the mice were killed four weeks later and liver tissues were collected.HE staining was used to observe the damage of liver tissue.Oil red O staining was used to observe the number of lipid droplets in liver tissue.The expression level of PGC-1αin liver was determined by IHC and Western Blot.The results showed that compared with the control group,the liver was not damaged after ligustrazine hydrochloride was added,the number of lipid drops was increased,and the expression level of PGC-1αwas significantly increased.In conclusion,ligustrazine hydrochloride could increase the expression of PGC-1αand promote liver heat production in mice under cold exposure.
