摘 要:目的探究骨关节炎(OA)滑膜中磷脂酶Cδ3(PLCD3)mRNA的表达情况及其与免疫细胞浸润的关系。方法基于前期发现的OA差异表达基因,通过收集临床OA组和对照组的滑膜组织,检测相关差异表达基因PLCD3 mRNA的表达差异,并采用CIBERSORT算法分析免疫细胞在OA组及对照组中的浸润模式,并进一步对PLCD3与浸润性免疫细胞的相关性进行分析。结果相较于对照组,PLCD3 mRNA相对表达水平在OA组滑膜组织中明显升高(P<0.05),OA滑膜组织中幼稚B细胞、活化NK细胞、M2巨噬细胞和活化肥大细胞的比例相对较高(P<0.05),PLCD3与这4种免疫细胞比例呈正相关(P<0.05)。结论PLCD3可能是诊断OA的关键生物标志物,其可能通过与浸润性免疫细胞的相互作用参与了OA的发病过程。Objective To investigate the expression of PLCD3 mRNA in the synovium of osteoarthritis(OA)and its relationship with immune cell infiltration.Methods Based on the differentially expressed genes of OA found in the previous study,the expression of phospholipase Cδ3(PLCD3)mRNA was detected by collecting synovial samples from OA group and control group.CIBERSORT algorithm was used to analyze the infiltration pattern of immune cells in OA group and control group,and the correlation between PLCD3 and infiltrating immune cells was further analyzed.Results Compared with the control group,the relative expression level of PLCD3 mRNA was significantly increased in synovial samples of OA group(P<0.05).The proportions of B cells naive,NK cells activated,M2 macrophages and mast cells activated in synovial tissues of OA group were relatively high(P<0.05).PLCD3 was positively correlated with the proportion of these four immune cells(P<0.05).Conclusion PLCD3 may be a key biomarker for the diagnosis of OA,which may be involved in the pathogenesis of OA by interacting with infiltrating immune cells.
