摘  要:目的 探究南柴胡地上部分不同极性部位抗菌作用及活性成分相关性,阐明抗菌药效物质。方法 采用不同浓度乙醇提取及体积分数为95%的乙醇溶液提取后用乙酸乙酯和正丁醇萃取得到10个极性部位,以金黄色葡萄球菌(Staphylococcus aureus)、大肠杆菌(Escherichia coli)、铜绿假单胞菌(Pseudomonas aeruginosa)、白色念珠菌(Candida albican)为对象,以药敏试验、最低抑菌浓度(minimun inhibitory concentration, MIC)、最小杀菌浓度(minimun bactericidal concentration, MBC)为指标,阐释抗菌活性部位。通过UPLC-Q-TOF-MS/MS对不同极性部位进行分析,确定共有成分,用双变量相关分析和灰色关联度分析,建立共有峰峰面积与MIC之间的相关性,筛选药效物质。结果 乙酸乙酯萃取部位为最佳抗菌部位。原儿茶酸、绿原酸、金丝桃苷、亚油酸、山奈酚、异槲皮苷、水仙苷、山奈素、柴胡色原酮酸9个成分与抗菌作用高度相关。结论 本研究为南柴胡地上部分在抗菌产品开发方面提供理论支撑。Objective To explore the antibacterial effect and the correlation of active components in the aboveground parts of Bupleurum scorzonerifolium Willd with different polarity, and to clarify the antibacterial substances.Method Ten polar parts of the aboveground part of B.scorzonerifolium Willd were obtained by ethanol extraction with different concentrations and 95% ethanol extraction with ethyl acetate and n-butanol.Staphylococcus aureus,Escherichia coli,Pseudomonas aeruginosa and Candida albican were used as the object, then the drug susceptibility test, minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) were determined to elucidate the best antibacterial polar site.Analysis of different polar sites by UPLC-Q-TOF-MS/MS technique to determine the consensus components, bivariate correlation analysis and Grey′s correlation analysis were used to establish the correlation between the peak area of the consensus and the level of MIC,and to screen the pharmacodynamic substances.Result Ethyl acetate extraction site is the best antibacterial site.Nine components, including protocatechuic acid, chlorogenic acid, hyperoside, linoleic acid, kaempferol, isoquercitrin, narcissoside, kaempferide, and saikochromic-acid, are highly associated with antibacterial effects.Conclusion This study provides theoretical support for the development of antibacterial products in the aboveground part of B.scorzonerifolium Willd.
