摘 要:目的探讨施旺细胞S-100蛋白在坐骨神经损伤大鼠阿司匹林修复再生中的作用机制。方法将新疆医科大学动物实验中心购入的120只雄性健康SD大鼠作为实验对象,按大鼠体重由轻至重编号,根据随机数字表法分成假手术组、模型组、阿司匹林组、阿司匹林+康复组,每组各30只。假手术组大鼠仅使坐骨神经暴露,模型组、阿司匹林组、阿司匹林+康复组大鼠制备坐骨神经损伤模型。假手术组和模型组在造模术毕后当日,取等体积0.9%氯化钠溶液进行灌胃,1次/d;阿司匹林组在造模术毕后当日,取1 mL阿司匹林溶液对大鼠行灌胃处理,1次/d;阿司匹林+康复组的灌胃方式与阿司匹林组相同,在术后1周拆线,之后接受康复训练。分别在术后4、6、8周检测各组大鼠的热缩足潜伏期(PWL)与机械性缩足反射阈值(MWT),并在术后8周对各组大鼠进行解剖与处死,检测大鼠的患侧与健侧腓肠肌湿质量比、感觉神经再生距离与施旺细胞S-100蛋白表达情况,分析S-100蛋白表达与各指标间的相关性。结果模型组4、6、8周的PWL和MWT均明显低于假手术组、阿司匹林组和阿司匹林+康复组,阿司匹林组、阿司匹林+康复组大鼠术后4、6、8周的PWL和MWT均明显低于假手术组,且阿司匹林+康复组各时段的PWL和MWT均明显高于阿司匹林组,差异均有统计学意义(P<0.05)。模型组的患侧与健侧腓肠肌湿质量比明显低于假手术组、阿司匹林组和阿司匹林+康复组,阿司匹林组、阿司匹林+康复组的患侧与健侧腓肠肌湿质量比明显低于假手术组,且阿司匹林+康复组明显高于阿司匹林组,差异均有统计学意义(P<0.05);模型组的感觉神经再生距离明显低于假手术组、阿司匹林组和阿司匹林+康复组,阿司匹林组、阿司匹林+康复组的感觉神经再生距离明显高于假手术组,差异均有统计学意义(P<0.05),但阿司匹林组与阿司匹林+康复组间比较,Objective To investigate the mechanism of Schwann cell S-100 protein in aspirin repair and regeneration in sciatic nerve injury rats.Methods A total of 120 male healthy SD rats purchased from Animal Experimental Center of Xinjiang Medical University were used as experimental subjects.The rats were numbered from light to heavy weight and divided into sham surgery group,model group,aspirin group,aspirin+rehabilitation group according to random number table method,with 30 rats in each group.The sham surgery group rats only exposed the sciatic nerve,while the model group,aspirin group,and aspirin+rehabilitation group rats prepared sciatic nerve injury models.On the day after the completion of the modeling surgery,the sham surgery group and model group were given an equal volume of 0.9%sodium chloride solution by gavage once a day;on the day after the completion of modeling,1 mL of aspirin solution was taken from the aspirin group and administered to the rats by gavage once a day;the gavage method of aspirin was the same as that of the aspirin+rehabilitation group,with stitches removed one week after surgery and rehabilitation training received afterwards.The paw withdrawal latency(PWL)and mechanical withdrawal threshold(MWT)of the rats were detected at 4,6 and 8 weeks after surgery,and the rats were dissected and killed at 8 weeks after surgery.The wet mass ratio of the gastrecnemius muscle on the affected side and the healthy side,the distance of sensory nerve regeneration and S-100 protein expression in Schwann cells were detected.The correlation between S-100 protein expression and each index was analyzed.Results The PWL and MWT of the model group at 4,6,and 8 weeks after surgery were significantly lower than those of the sham surgery group,the aspirin group and the aspirin+rehabilitation group,the PWL and MWT of the aspirin group and the aspirin+rehabilitation group were significantly lower than those of the sham surgery group at 4,6,and 8 weeks after surgery,and the PWL and MWT of the aspirin+rehabilitation grou
