摘  要:目的:研究柴胡疏肝散加味江香薷对肥胖抑郁大鼠的作用。方法:造抑郁症模型后30 d将大鼠分为肥胖抑郁模型组、柴胡疏肝散加味江香薷10、20、40 g/kg组、氟西汀1.8 mg/kg组,各组灌胃相应药物或生理盐水,给药30 d后检测抑郁大鼠在强迫游泳试验中的指标及糖水偏好率、血脂和炎症因子含量;大鼠粪便进行16S rDNA高通量测序技术获取数据进行分析。结果:与正常对照组比较,肥胖抑郁模型TC、TG、LDL-C含量明显升高(P<0.05或P<0.01),糖水偏好率显著降低,游泳总距离、直立次数降低,游泳静止时间显著增加(P<0.01),血清IL-1β、IL-6、TNF-α、LPS含量明显升高(P<0.05或P<0.01),结肠结构发生病理改变;与肥胖抑郁模型比较,柴胡疏肝散化裁方加味江香薷40 g/kg组大鼠行为学改善,糖水偏好率提高(P<0.05),各给药组大鼠旷场行为中直立次数和运动总距离明显增加(P<0.05或P<0.01),强迫游泳静止时间明显降低P<0.05),血脂含量明显降低(P<0.05),IL-1β、IL-6、TNF-α、IL-27、LPS含量明显降低(P<0.05);肠道菌群的丰富度增加且肠道菌群组间差异具有显著性(P<0.05),同时能显著影响肠道革兰氏阳性和阴性菌的来源和比例。结论:柴胡疏肝散加味江香薷可能通过调节肠道菌群的相对丰度和种类来达到治疗肥胖抑郁症的效果。Objective:To investigate the effects of Chaihu Shugan Powder(柴胡疏肝散)modified with Jiangxiangru(江香薷)on obese and depressed rats.Methods:After 30 days of depression model induction,model rats were divided into a model group,Chaihu Shugan Powder modified with Jiangxiangru groups(10,20,and 40 g/kg),and a fluoxetine group(1.8 mg/kg).The corresponding drugs or saline were administered orally for 30 days.After 30 days of treatment,forced swimming test,rate of sucrose water preference,lood lipid levels and inflammatory factor levels were measured.Fecal samples were collected from the rats for high-throughput sequencing of 16S rDNA,and the data were analyzed.Results:Compared with the control group,the model group elevated the levels of TC,TG,LDL-C(P<0.05 or P<0.01),decreased preference for sucrose water,total distance traveled and standing times,increased immobility time in the forced swimming test(P<0.01),and increased the levels of IL-1β,IL-6,TNF-α,IL-27,and LPS(P<0.05 or P<0.01),and the pathological changes of colon structure were observed.Compared with the model group,the Chaihu Shugan Powder modified with Jiangxiangru group at 40 g/kg showed improved behavioral performance and increased preference for sucrose water(P<0.05),and the groups with drug treatment showed increased standing times and total distance traveled in the open field test(P<0.05 or P<0.01),decreased immobility time in the forced swimming test(P<0.05),decreased blood lipid levels(P<0.05),and decreased levels of IL-1β,IL-6,TNF-α,IL-27,and LPS(P<0.05).The abundance of the gut microbiota increased,and there were significant differences in the composition of the gut microbiota among the groups(P<0.05).Moreover,the treatment significantly affected the source and proportion of Gram-positive and Gram-negative bacteria in the gut.Conclusion:Chaihu Shugan Powder modified with Jiangxiangru may achieve therapeutic effects on obesity and depression by regulating the relative abundance and composition of the gut microbiota.
