香连丸及有效单体黄连素联合5-氟尿嘧啶对HCT116结肠癌荷瘤裸鼠的影响

摘　　要：目的通过建立结肠癌荷瘤裸鼠模型和结肠癌HCT116细胞实验,探讨香连丸及有效单体黄连素联合5-FU对结肠癌HCT116细胞的抑制作用。方法(1)建立HCT116结肠癌荷瘤裸鼠模型,给予香连丸,黄连素,5-氟尿嘧啶,香连丸和黄连素+5-FU干预,观察各组药物对肿瘤大小影响。PCR法检测裸鼠瘤体组织中Caspase-3,TNF,TP53 mRNA的表达;(2)建PCR法检测裸鼠瘤体组织中Caspase-3,肿瘤坏死因子(tumor necrosis factor,TNF),TP53 mRNA表达;和检测体外培养的HCT116细胞中Caspase-3,TNF,TP53 mRNA表达;(3)CCK8法观察黄连素组、5-FU组、黄连素+5-FU联合用药组对HCT116细胞增殖的影响。结果(1)通过建立结肠癌荷瘤裸鼠模型,与模型对照组比较,香连丸组、黄连素组、香连丸+5-FU联合用药组和黄连素+5-FU联合用药组的肿瘤瘤体均有所减小,抑瘤率增加,差异有统计学意义(P<0.05);与香连丸、5-FU和黄连素单药组比较,黄连素+5-FU联合用药组的裸鼠瘤体明显减小,抑瘤率显著增加,差异有统计学意义(P<0.01)。(2)体内实验的PCR结果显示:与模型对照组比较,香连丸组、黄连素组、5-FU组、香连丸+5-FU联合用药组和黄连素+5-FU联合用药组Caspase-3表达均没有上升,差异均无统计学意义(P>0.05)。与模型对照组比较,黄连素组TNF表达明显上升,差异有统计学意义(P<0.01);与模型对照组比较,香连丸+5-FU联合用药组、黄连素+氟尿嘧啶联合用药组TNF表达上升,但差异无统计学意义(P>0.05);与模型对照组比较,5-FU组TNF表达无明显上升,且差异无统计学意义(P>0.05);与香连丸、5-FU单药组比较,香连丸+5-FU联合用药组TNF表达明显上升,且差异有统计学意义(P<0.05);与模型对照组比较,黄连素组TP53表达明显上升,差异有统计学意义(P<0.01);与模型对照组比较,5-FU和香连丸单药组TP53表达下降,差异无统计学意义(P>0.05);与香连丸单药组比,香连丸+5-FU联合用药组、黄连素+5-FU联合用药组Objective To explore the inhibitory effect of Xianglian Pills or berberine combined with 5-fluorouracil on colon cancer HCT116 cells based on the experiments with colon cancer tumor-bearing nude mice and colon cancer HCT116 cells.Methods(1)The HCT116 colon cancer tumor-bearing nude mouse model was established and treated with Xianglian Pills,berberine,5-fluorouracil,Xianglian Pills+5-fluorouracil,and berberine+5-fluorouracil,respectively,and the tumor size in each group was observed.(2)PCR was employed to determine the mRNA levels of caspase-3,tumor necrosis factor(TNF),and TP53 in the tumor tissue and the HCT116 cells cultured in vitro.(3)The CCK8 assay was employed to examine the proliferation of HCT116 cells in the berberine group,5-fluorouracil group,and berberine+5-fluorouracil group.Results(1)In the animal experiments,compared with the model control group,Xianglian Pills,berberine,Xianglian Pills+5-fluorouracil,and berberine+5-fluorouracil all decreased the tumor size,showing tumor-inhibiting effects(P<0.05).Compared with Xianglian Pills,5-fluorouracil,and berberine alone,berberine+5-fluorouracil reduced the tumor size and showed an increased tumor inhibition rate(P<0.01).(2)PCR results of animal experiments:Compared with the model control group,Xianglian Pills,berberine,5-fluorouracil,Xianglian Pills+5-fluorouracil,and berberine+5-fluorouracil did not up-regulate the expression of caspase-3(all P>0.05).Compared with the model control group,berberine up-regulated the expression of TNF(P<0.01),while Xianglian Pills+5-fluorouracil and berberine+5-fluorouracil slightly up-regulated the expression of TNF(P>0.05).Compared with Xianglian Pills and 5-fluorouracil alone,Xianglian Pills+5-fluorouracil up-regulated the expression of TNF(P<0.05).Compared with the model control group,berberine down-regulated the expression of TP53(P<0.01),while 5-fluorouracil and Xianglian Pills alone only slightly down-regulated the expression of TP53(P>0.05).Compared with Xianglian Pills alone,Xianglian Pills+5-fluorouracil,berberine
