摘 要:目的分析当归挥发油对人结直肠癌(Colorectal cancer,CRC)细胞HCT116生物学行为的影响作用。方法人CRC HCT116细胞按照0μg/ml、25μg/ml、50μg/ml、100μg/ml 4个当归挥发油浓度分为对照组、当归挥发油25μg/ml组、50μg/ml组、100μg/ml组。MTT、CCK-8实验检测细胞增殖情况;Transwell实验检验细胞迁移、侵袭能力;流式细胞术检测CRC HCT116细胞凋亡率;Western bolt检测各组细胞凋亡蛋白B细胞淋巴瘤-2(B-cell lymphoma-2,Bcl-2)和Bcl-2相关X蛋白(Recombinant Bcl-2 associated X protein,Bax)变化情况。结果MTT实验结果显示,当归挥发油25μg/ml、50μg/ml、100μg/ml组细胞增殖抑制率均明显高于对照组,且HCT116细胞增殖抑制率随当归挥发油剂量的上升而上升,差异有统计学意义(P<0.05);CCK-8实验结果显示,三组细胞增殖计数均明显低于对照组,且HCT116细胞增殖率随当归挥发油剂量的上升而下降,差异有统计学意义(P<0.05);Transwell实验结果显示对照组的细胞迁移、侵袭个数明显高于当归挥发油25μg/ml、50μg/ml、100μg/ml组,差异有统计学意义(P<0.05);流式细胞术检测HCT116凋亡结果显示相比于对照组,当归挥发油25μg/ml、50μg/ml、100μg/ml组的HCT116细胞凋亡率明显升高,差异有统计学意义(P<0.05);Western bolt结果显示,与对照组比较,当归挥发油各组HCT116细胞的Bax蛋白相对表达量均明显升高,Bcl-2蛋白相对表达量均明显降低,差异有统计学意义(P<0.05)。结论当归挥发油能够显著抑制人CRC HCT116细胞增殖、迁移等细胞生物学行为,同时能够提高细胞凋亡率,其可能通过调控Wnt/β-catenin通路参与细胞凋亡过程。Objective To analyze the effects of volatile oil of Angelicae Sinensis Radix on the behaviors of the human colorectal cancer(CRC)cell line HCT116.Methods The HCT116 cells were classified into a control group and volatile oil(25,50,and 100μg/mL)groups.The cell proliferation was examined by MTT and CCK-8 assays.Transwell assay was employed to examine the cell migration and invasion.The apoptosis rate of HCT116 cells was detected by flow cytometry.Western blotting was employed to determine the expression levels of the apoptosis-associated proteins B-cell lymphoma-2(Bcl-2)and recombinant Bcl-2-associated X protein(Bax)in each group.Results Compared with the control group,the volatile oil of Angelicae Sinensis Radix inhibited the proliferation of HCT116 cells in a dose-dependent manner(P<0.05),and the proliferation rate of HCT116 cells decreased with the increase in the dose of the volatile oil(P<0.05).The number of cell migration and invasion in the control group was higher than that in the volatile oil groups(P<0.05).Compared with the control group,the volatile oil of Angelicae Sinensis Radix at different doses increased the apoptosis rate of HCT116 cells(P<0.05).Moreover,the volatile oil up-regulated the relative protein level of Bax and downregulated the relative protein level of Bcl-2 in HCT116 cells(P<0.05).Conclusion The volatile oil of Angelicae Sinensis Radix can significantly inhibit the proliferation and migration and increase the apoptosis rate of HCT116 cells,and it may participate in the apoptosis by regulating the Wnt/β-catenin pathway.
