摘 要:过氧化氢酶在植物生理生化过程中具有重要的作用,为了简便、快速、准确地测定植物过氧化氢酶活性,进行了紫外分光光度法测定烟苗叶片过氧化氢酶活性的研究。结果表明,利用紫外分光光度法测定植物过氧化氢酶活性所需试剂种类少、操作简单、测定快速。标准曲线相关性好,样品测定重现性好,加标回收率高,测定结果与高锰酸钾滴定法呈极显著相关,说明方法准确可靠。本方法稳定时间至少在5 h以上,稳定时间长有利于大批量的样品分析测定。利用紫外分光光度法标定过氧化氢浓度与高锰酸钾滴定法无显著差异,说明用紫外分光光度法直接标定过氧化氢浓度可行,既可简化操作和节省时间,又能降低分析测试成本。因此本研究为植物过氧化氢酶的研究提供了一种简便快速、稳定可靠、成本低廉的分析测定方法。对两种烟苗叶片的测定表明,楚雪26烟苗叶片的过氧化氢酶活性高于楚雪14,烟苗叶片过氧化氢酶活性呈上半叶>全叶>下半叶,说明烟苗叶片过氧化氢酶活性受不同烟苗品种影响,且过氧化氢酶在烟苗叶片分布并不均匀。Catalase plays an important role in plant biochemical and physiological process.A study of determining plant catalase activity by ultraviolet spectrophotometry was conducted to explore an easy,quick and accurate method of determining plant catalase activity.The results indicated that the use of ultraviolet spectrophotometry to determine plant catalase activity required fewer types of reagents,simple operation,and had fast determination.This method was accurate and reliable because of the high linear correlation of standard curve,good reproducibility,high recovery rate and the extremely significant correlation between the determination of catalase activities by the method of the ultraviolet spectrophotometry and potassium permanganate titration.The stable time of this method was at least 5 hours,and the long stable time was suitable to the assay of large quantity samples.With no significant difference of calibration of hydrogen peroxide concentration between the ultraviolet spectrophotometry and potassium permanganate titration,it was practicable to calibrate the hydrogen peroxide concentration by the ultraviolet spectrophotometry,and this method can not only simplify operation and save times,but also reduce analysis costs.In a word,this study provides a easy,quick,stable,reliable and low cost method for determining plant catalase activity.The results of determining the catalase activity of two species of tobacco seedling leaves showed that the catalase activity was higher in Chuxue 26 than that of in Chuxue 14,and was in the order of upper half leaf>whole leaf>lower half leaf.This indicated that the catalase activity in tobacco seedling leaves was affected by the species of tobacco seedling and the distribution of catalase in tobacco leaves was uneven.
