摘 要:目的探讨新型双组分交联封闭聚合物水凝胶对大鼠创面愈合的作用。方法采用背部皮肤打孔法构建SD大鼠创面模型,再按随机数字表法分为水凝胶组、硫酸庆大霉素组、对照组,每组6只。水凝胶组每天在创面处涂抹植物纤维素衍生物水溶液25μL,同时喷涂氧化淀粉水溶液25μL;硫酸庆大霉素组每天在创面处涂抹硫酸庆大霉素50μL;对照组不作任何处理。药物干预期间记录每天创面愈合情况并测量创面面积;药物干预10 d后麻醉大鼠,取创面组织测量创面肉芽组织面积,并检测组织中IL-6、TNF-α、转化生长因子β(TGF-β)mRNA相对表达量。结果从药物干预第4天起,水凝胶组和硫酸庆大霉素组大鼠创面面积均明显小于对照组(均P<0.05),而水凝胶组与硫酸庆大霉素组比较差异均无统计学意义(均P>0.05)。水凝胶组、硫酸庆大霉素组创面肉芽组织面积均明显小于对照组,组织中IL-6、TNF-αmRNA相对表达量均明显低于对照组,TGF-βmRNA相对表达量明显高于对照组,差异均有统计学意义(均P<0.05);而水凝胶组与硫酸庆大霉素组创面肉芽组织面积以及组织中IL-6、TNF-α、TGF-βmRNA相对表达量比较,差异均无统计学意义(均P>0.05)。结论新型双组分交联封闭聚合物水凝胶可加速大鼠创面愈合,减少炎症反应。Objective To explore the impact of a novel dual-component crosslinked encapsulated polymer hydrogel in wound healing of rats.Methods A wound model was developed by piercing the dorsal skin in SD rats.The rats were then randomly divided into three groups,namely the hydrogel group,gentamycin sulfate group and control group,with 6 rats in each group.The hydrogel group was treated daily with 25μL aqueous solution of plant cellulose derivative on the wound site,along with the application of 25μL oxidized starch solution.The gentamicin sulfate group was treated with 50μL gentamycin sulfate on the wound site every day.The control group received no treatment.The wound healing conditions were observed and the wound surface area was measured during drug intervention.After 10 days of drug intervention,the rats were anesthetized and the area of granulation tissue in the wound site was measured,and the relative expression levels of IL-6,TNF-αand transforming growth factorβ(TGF-β)mRNA in the tissues were detected.Results From the fourth day of drug intervention,the wound surface area of rats in the hydrogel group and the gentamicin sulfate group was both significantly smaller than that of the control group(both P<0.05).However,there was no statistical significance in wound surface area between the hydrogel group and the gentamycin sulfate group at all time-points(all P>0.05).In the hydrogel and gentamycin sulfate groups,areas of the granulation tissues were both significantly smaller than those of the control group,the expression levels of IL-6 and TNF-αmRNA in the tissues were significantly lower,and TGF-βmRNA expression level was significantly higher(all P<0.05).No statistically significant differences in area of wound granulation tissues,and relative expression levels of IL-6,TNF-αand TGF-βmRNA were observed between the hydrogel group and the gentamicin sulfate group(all P>0.05).Conclusion This new dual-component crosslinked encapsulated polymer hydrogel accelerates wound healing and reduces inflammation of rats.
