健脾复方对人大肠癌移植瘤裸小鼠不同组织AP-1表达的影响

摘　　要：目的探索人大肠癌移植瘤裸小鼠肿瘤组织、肿瘤邻近正常肠组织和远端正常肠组织中激活蛋白-1(activator protein-1,AP-1)表达的差异以及健脾复方对这些AP-1表达的影响。方法将40只BALB/C裸小鼠按1∶1∶1∶1随机分入健脾复方组、5氟尿嘧啶(5-flurouracil,5-Fu)组、模型对照组和空白对照组,前3组建立人大肠癌原位移植瘤模型,分组治疗8周。采用免疫组化法检测各组肿瘤组织、肿瘤邻近(距肿瘤切缘1 cm)正常肠组织和远端(距肿瘤切缘>3 cm)正常肠组织的AP-1表达。结果模型对照组肿瘤组织和肿瘤邻近正常肠组织的AP-1表达高于空白对照组及远端正常肠组织;健脾复方组、5-Fu组和模型对照组的远端正常肠组织中也观察到AP-1平均光密度增高。健脾复方组肿瘤组织、肿瘤邻近正常肠组织AP-1表达与空白对照组以及远端正常肠组织未见显著差异,并有低于模型对照组的趋势,在5-Fu组可以看到类似的结果。结论肿瘤组织、肿瘤邻近正常肠组织甚至远端正常肠组织AP-1的异常升高与肿瘤生长、侵袭和转移存在一定关系,健脾复方的治疗在一定程度上能降低肿瘤组织和肿瘤邻近正常肠组织的AP-1表达,但其作用机制仍有待进一步研究。Objective To investigate differences of activator protein-1(AP-1)expression among tumor tissue,adjacent normal colon tissue and distal normal colon tissue in nude mice with human colorectal cancer grafts and impact of Jianpi Compound Recipe(健脾复方)on these expressions.Methods Forty BALB/C nude mice were randomized into Jianpi Compound Recipe group,5-fluorouracil(5-Fu)group,model control group and blank control group.Human colorectal cancer orthotopic transplanted tumor model was established in the first 3 groups.All groups received respective treatments for 8 weeks.Specimens were incised from tumor tissues,adjacent(1 cm distance from tumor tissue)normal colon tissues,and distal(>3 cm distance from tumor tissue)normal colon tissues.The expressions of AP-1 were assessed by immunohistochemistry method.Results AP-1 expression in tumor tissue and adjacent normal colon tissue was higher in model control group than that in blank control group and distal normal colon tissue.Mean optical density of AP-1 was also higher in Jianpi Compound Recipe group,5-Fu group and model control group.AP-1 expression in tumor tissue and adjacent normal colon tissue in Jianpi Compound Recipe group showed non-significant difference with blank control group and distal normal colon tissue,and showed lower trends than that in model control group.Similar results were also showed in 5-Fu group.Conclusion Higher expression of AP-1 in tumor tissue,adjacent colon tissue and even in distal normal colon tissue has relationship with tumor growth,invasion and metastasis.Jianpi Compound Recipe may have potent to reduce the AP-1 expression,but the mechanism of action need further research.
