柏叶加味香波对银屑病样小鼠模型作用的研究

摘　　要：目的研究柏叶加味香波对咪喹莫特诱导的银屑病小鼠模型的影响。方法将柏叶加味香波应用于银屑病样小鼠模型。BALB/c小鼠60只随机分成6组,空白基质组(CON组),模型组[咪喹莫特乳膏(imiquimod,IMQ)组],乐银洗发液组(LY组),柏叶加味香波低(C-L组)、中(C-M组)、高(C-H组)浓度组。使用5%咪喹莫特诱导银屑病样小鼠模型。CON组、IMQ组均匀涂抹无药香波基质,其他用药组外用给药保留15 min后洗去,1次/d,连续8 d给药。观察小鼠一般情况及体质量趋势、计算脏器指数、背部皮损进行银屑病面积和严重度指数(psoriasis area and severity index,PASI)评分、观察组织病理学形态改变、使用实时荧光定量聚合酶链反应(qRT-PCR)方法检测皮损组织中的白细胞介素-17A(interleukin-17A,IL-17A)及白细胞介素-23(interleukin-23,IL-23)的相对表达量,采用单因素ANOVA方差分析进行统计学分析。结果给药8 d后小鼠体质量不同程度下降,与IMQ组相比,LY、C-L、C-M、C-H组小鼠体质量较IMQ组重,背部银屑病样皮损改善,银屑病面积和严重度指数(psoriasis area and severity index,PASI)评分均下降(P<0.01);与CON组相比,胸腺指数下降,而其余各组脾脏指数升高。LY组与CH组脾脏指数小于IMQ组,用药一定程度上缓解炎症反应;与CON组相比,其余几组小鼠组织病理学形态都有所改变,与IMQ组相比,LY、C-L、C-M、C-H组表皮增生情况减轻,厚度变薄,CH、CM、LY组纤维组织增生程度减轻;与IMQ组相比,LY、C-L、C-M、C-H组小鼠皮肤组织中的IL-17A mRNA及IL-23 mRNA相对表达量均明显下降,差异有统计学意义(P<0.01),其中C-H组与LY组下降最明显。结论柏叶加味香波可以通过阻断IL-23/IL-17-Th17轴的活化减少IL-17A、IL-23的表达、表皮层角质细胞的增生,降低小鼠脾脏指数,以改善咪喹莫特诱导的小鼠皮损严重程度,表明此香波可达到一定调理头皮银屑病的作用。Objective To study the effect of Modified Cypress Leaves Shampoo on imiquimod-induced psoriasis mouse model.Methods Modified Cypress Leaves Shampoo was applied to psoriasis-like mouse model.Sixty BALB/C mice were randomly divided into 6 groups,blank matrix group(CON group),model group[imiquimod(IMQ)group],Leyin shampoo group(LY group),cypress leave-flavored shampoo group with low concentration(C-L group),medium concentration(C-M group)and high concentration(C-H group).Psoriasis-like mouse models were induced with 5%imiquimod.CON group and IMQ group were evenly applied with drug-free shampoo matrix,and the other drug groups were reserved for external administration for 15 minutes and then washed off,once a day,for consecutive 8 days.The general condition and body weight trend of mice were observed,organ index was calculated,psoriasis area and severity index(PASI)score was performed on skin lesions of the back,histopathological morphological changes were observed,the relative expression levels of interleukin-17A(IL-17A)and interleukin-23(IL-23)in skin lesions were detected by qRT-PCR,and statistical analysis was performed by one-way ANOVA.Results After 8 days of administration,the body weight of LY,C-L,C-M and C-H mice was heavier than that of IMQ group,the psoriatic skin lesions on the back were improved and the PASI score was decreased(P<0.01).Compared with those of the CON group,thymus index decreased,while spleen index increased in other groups.The spleen indexes of LY group and CH group were lower than that of IMQ group.Compared with that of the CON group,the histopathological morphology of the other groups was changed.Compared with IMQ group,the epidermal hyperplasia was reduced in LY,C-L,C-M and C-H groups,and the degree of fibrous tissue hyperplasia was reduced in CH,CM and LY groups.Compared with those of the IMQ group,the relative expression levels of IL-17A mRNA and IL-23 mRNA in LY,C-L,C-M and C-H mice skin tissue were significantly decreased,with statistically significant differences(P<0.01),among whic
