摘  要:目的通过观察二陈汤对脑出血急性期大鼠胃肠组织干细胞因子(stem cell factor,SCF)/干细胞因子受体(C-kit)信号通路的动态变化的影响,探讨二陈汤对脑出血急性期胃肠动力障碍的作用机制及与脑出血的关系。方法将120只大鼠随机均分为假手术组、模型组、西药组和中药组,采用自体血注入法制备大鼠脑出血模型。假手术组、模型组给予10 mL/(kg·d)生理盐水、西药组和中药组分别给予10 mL/(kg·d)的莫沙必利悬浊液、二陈汤悬浊液灌胃,分别于给药1 d、4 d、7 d处死大鼠,采用RT-PCR法和Western Blot法检测大鼠胃、十二指肠组织中的SCF、C-kit蛋白及mRNA的表达水平。结果模型组大鼠胃肠组织中SCF、C-kit mRNA表达较假手术组显著降低(P<0.05),且随时间延长逐渐降低,于实验结束(7 d)时达到最低水平;西药组、中药组大鼠胃肠组织中SCF、C-kit mRNA表达水平较模型组显著升高(P<0.05),且随时间推移逐渐升高,大鼠胃肠SCF、C-kit mRNA表达水平均于实验结束时达到最高水平;中药组大鼠胃肠组织中SCF、C-kit mRNA表达水平在7 d的实验中均高于西药组,二者相比有明显统计学意义(P<0.05)。结论脑出血急性期胃肠动力障碍的作用机制与SCF/C-kit信号通路相关蛋白及mRNA的表达降低有关,通过积极干预,胃肠功能障碍可以得到恢复。Objective To observe the effect of Erchen Decoction(二陈汤)on the dynamic changes of stem cell factor(SCF)/c-kit signal pathway in gastrointestinal tissue of rats with acute intracerebral hemorrhage and explore the mechanism of Erchen Decoction on gastrointestinal motility disorder in acute intracerebral hemorrhage and its relationship with intracerebral hemorrhage.Methods A total of 120 rats were randomly divided into sham operation group,model group,western medicine group and TCM group.The sham operation group and the model group were given 10 mL/(kg·d)normal saline,the western medicine group and the TCM group were given 10 mL/(kg·d)mosapride suspension and Erchen Decoction suspension by gavage respectively.The rats were killed on the 1st,4th and 7th day of administration.The expression levels of SCF,c-kit protein and mRNA in rat stomach and duodenum were detected by RT-PCR and Western blot.Results The expressions of SCF and c-kit mRNA in gastrointestinal tissue of the model group were significantly lower than those in the sham operation group(P<0.05),and gradually decreased with the time,reaching the lowest level at the end of the experiment(7 days).The expression levels of SCF and c-kit mRNA in gastrointestinal tissues of rats in the western medicine group and TCM group were significantly higher than those in the model group(P<0.05),and gradually increased with the time.The expression levels of SCF and c-kit mRNA in gastrointestinal tissues of rats reached the highest level at the end of the experiment.The expression levels of SCF and c-kit mRNA in gastrointestinal tissue of rats in the TCM group were significantly higher than those in the western medicine group(P<0.05).Conclusion The mechanism of gastrointestinal motility disorder in the acute stage of intracerebral hemorrhage is related to the decreased expression of SCF/c-kit signal pathway related proteins and mRNA,which can be recovered by active intervention of gastrointestinal dysfunction.
