胃肠安通过RUFY3 抑制胃癌腹膜转移的机制研究

摘　　要：目的研究健脾复方胃肠安对人胃癌细胞裸鼠腹膜转移的影响及其作用机制。方法构建裸鼠腹膜转移模型,随机分为空白组、模型组、胃肠安组和五氟脲嘧啶(5-fluorouracil,5-Fu)组,不同干预后,观察各组腹腔结节、肠系膜结节和肝转移结节数量。免疫组化比较各组腹腔肿瘤组织侵袭迁移相关蛋白表达情况,酶联免疫吸附测定法(enzyme linked immunosorbent assay,ELISA)检测血清中侵袭迁移相关标志物。结果胃肠安干预后,腹腔转移结节、肠转移结节和肝转移结节数量均明显下降,肿瘤相对较小,部分肝转移瘤内可见炎细胞浸润。与模型组相比,胃肠安显著降低了血清中基质金属蛋白酶2(matrix metalloproteinase 2,MMP2)、基质金属蛋白酶9(matrix metalloproteinase 9,MMP9)及血管内皮生长因子(vascular endothelial growth factor,VEGF)的表达。此外,胃肠安抑制了腹腔瘤体组织RUN和FYVE结构域蛋白3(RUN and FYVE domain protein 3,RUFY3)、MMP2、MMP9及VEGF蛋白的表达水平。结论胃肠安通过调控RUFY3、MMPs以及VEGF等蛋白抑制胃癌腹膜转移。Objective To study the mechanism of spleen-strengthening traditional Chinese medicine(TCM)prescriptionWeichang′an(胃肠安,WCA)on peritoneal metastasis of human gastric cancer cells in nude mice.Methods The model of peritoneal metastasis in nude mice was established and randomly divided into blank group,model group,Weichang′an group and 5-fluorouracil(5-Fu)group.After different intervention,the numbers of abdominal nodules,mesenteric nodules and liver nodules in each group were counted.Immunohistochemistry method was used to compare the expressions of invasion and migrationrelated proteins in abdominal tumor tissues of each group,and enzymelinked immunosorbent assay(ELISA)was used to detect the invasion and migrationrelated markers in serum.Results After the intervention of Weichang′an,the numbers of abdominal nodules,intestinal nodules and liver nodules decreased significantly,the tumors were relatively smalland the inflammatory cell infiltration was found in some liver metastases.Compared with those of the model group,the expressionsof matrix metalloproteinase 2(MMP2),matrix metalloproteinase 9(MMP9)and vascular endothelial growth factor(VEGF)in serum were significantly decreased by Weichang′an,and the levels of RUN and FYVE domain protein 3(RUFY3),MMP2,MMP9 and VEGF in abdominal tumor tissues were also inhibited.Conclusion WCA inhibits peritoneal metastasis of gastric cancer by regulating RUFY3,MMPs and VEGF.
