摘 要:【目的】了解云南省猪场捷申病毒(porcine teschovirus,PTV)的感染情况及流行毒株分子进化特征,为预防控制该病提供参考。【方法】从云南曲靖、弥勒和大理的猪场随机采集粪便样品,采用RT-PCR方法对PTV 5′-UTR部分基因片段进行扩增、克隆及测序,通过生物信息学软件进行同源性分析并构建系统进化树。【结果】PTV核酸阳性率达27.3%(32/117),测序后去除重复序列得到6条序列。同源性分析结果显示:株间核苷酸同源性为93.2%~98.9%,与参考序列核苷酸同源性为89.7%~100.0%;系统进化树构建结果显示:YNDL-2和YNDL-3与四川省分离的PTV毒株亲缘关系较近;YNQJ-1与日本分离的PTV毒株亲缘关系较近;YNDL-1、YNQJ-4和YNQJ-5聚为一簇,与多米尼加分离的毒株亲缘关系较近,表明同一猪场存在不同血清型感染。【结论】云南猪场PTV感染率较高,流行株间进化关系差异较大,同一猪场存在不同基因型的病毒感染,未体现出明显的地域性。研究结果为PTV致病机理、疫苗研制及防控等奠定了基础。[Purpose]To understand the porcine teschovirus(PTV)infection and the molecular evolution characteristics of prevalence strains in Yunnan Province,thus lay foundation to the preven-tion and control of this disease in pig farms.[Methods]Fecal samples were randomly collected from pig farms in Qujing,Mile and Dali in Yunnan Province,and the partial gene fragment of PTV 5′-UTR was amplified,cloned and sequenced by RT-PCR.Homology analyzed by using bioinformat-ics software and a phylogenetic tree was constructed.[Results]The positive rate of PTV in collec-ted samples was 27.3%(32/117).After sequences comparing,repeated sequences were removed and six prevalence strain sequences finally achieved.Homology analysis results showed 93.2%-98.9%nucleotide identity between strains,and 89.7%-100.0%with selected reference sequence.The phylo-genetic tree showed that YNDL-2 and YNDL-3 strains were closely related to the PTV strains isol-ated in Sichuan Province;YNQJ-1 was closely related to Japanese strain;YNDL-1,YNQJ-4 and YN-QJ-5 clustered together and were closely related to the isolated strain in the Dominican Republic.It was indicated that there were different serotypes infection in the same pig farm.[Conclusion]PTV is highly prevalence in Yunnan pig farm,and great evolutionary relationship differences found in de-tected strains.In the same pig farm,there are at least two genotypes co-infection,and no obviously re-gional feature is found.Results lay a foundation for further study of pathogenesis,vaccine develop-ment and effective control of PTV.
